摘  要:目的:比较角叉菜λ-卡拉胶乙酰化修饰前后对细胞免疫活性和抗氧化活性的变化,探讨乙酰化修饰对角叉菜多糖λ-卡拉胶生物活性的影响。方法:从角叉菜中提取多糖分级得到λ-卡拉胶,并对其进行乙酰化修饰;采用MTT比色法对角叉菜λ-卡拉胶及其乙酰化衍生物c-λ-卡拉胶对小鼠淋巴细胞和巨噬细胞的免疫功能进行了体外试验和比较,采用化学方法产生羟自由基和小鼠肝匀浆液脂质过氧化体系,对λ-卡拉胶和c-λ-卡拉胶进行抗氧化活性检测。结果:λ-卡拉胶和c-λ-卡拉胶对无诱导的小鼠脾淋巴细胞均有促增殖作用,其中同浓度组c-λ-卡拉胶对小鼠脾淋巴细胞促增殖作用更强。与λ-卡拉胶相比,c-λ-卡拉胶对ConA和LPS诱导的脾淋巴细胞增殖反应均有较明显促进作用。c-λ-卡拉胶对单核巨噬细胞的吞噬功能的促进作用也强于λ-卡拉胶,乙酰化修饰可以显著增强角叉菜λ-卡拉胶对羟自由基和肝匀浆脂质过氧化的抑制作用。结论:角叉菜λ-卡拉胶的乙酰化修饰能有效的提高其免疫活性和抗氧化活性。Objective:To study the effect of acetylated modification on the biological activities of λ carrageenan,he immunomodulation and antioxidant activities of λ-carrageenan and it's acetylated derivative(c-λ-carrageenan) were investigated.Methods:The λ-carrageenan was obtained from Chondrus ocellatus and was acetylated by pyridine/acetic anhydride.MTT colorimetric analysis was used to evaluate the immunomodulation activities of spleen-lymphopoiesis and macrophage cell.Antioxidant activities of λ carrageenan and c-λ-carrageenan by chemical methods,to produce hroxide free radicals and lipid peroxide of liver homogenate.Results:λ-Carrageenan and c-λ-carrageenan can improve proliferation of spleen lymphocyte,and splenocytes induced by ConA or LPS.c-λ-carrageenan had significantly effect on proliferation of splenocytes than λ-carrageenan,additionally c-λ-carrageenan could enhance ConA or LPS induced splenocytes proliferation.Also c-λ-carrageenan could increase the phagocytic activity of mice peritoneal macrophages.Results:indicated that acetylated modification could significantly improve the scavenging capacity of hydroxyl radicals and the inhibiting of lipid peroxide of liver homogenate.Conclusion:These results suggest that acetylation can improve the immunomodulation activities and antioxidant activities of λ-carrageenan.
