摘 要:目的:建立一种耐药性稳定的人涎腺黏液表皮样癌细胞株(MEC/5-FU),为研究其耐药机制和逆转药物的筛选奠定实验基础。方法:采用浓度递增法联合大剂量冲击诱导方法,建立稳定的MEC/5-FU。应用MTT法、RT-PCR、罗丹明123蓄积试验等方法检测该细胞株的耐药性能及其生物学特性。结果:MEC/5-FU耐药性能稳定,对5-FU的耐药倍数达到36.11,并且对其他几种化疗药物表现不同程度的交叉耐药性。RT-PCR显示耐药相关基因ABCA2、ABCB1、ABCB11、ABCC1、ABCC4等ABC家族基因在MEC/5-FU细胞中明显高表达。罗丹明123在MEC/5-FU细胞中的蓄积量远低于MEC细胞。结论:MEC/5-FU具有多药耐药特性,为人涎腺黏液表皮样癌耐药机制和耐药逆转研究提供了理想的实验模型。Objective: To establish a multidrug resistant(MDR) cell line(MEC/5-FU) of human salivary gland mucoepidermoid carcinoma and to research its characterization.Methods: Human mucoepidermoid carcinoma cell line MEC was treated by pulse treatments of high concentration combining with continuous gradually increased concentration exposed to 5-fluorouracil(5-FU) to introduce its multidrug resistance(MEC/5-FU).The sensitivity of the multidrug resistance was measured by MTT assay.The expression of ABC transporters genes were analyzed by RT-PCR.Rhodamine 123 was used for functional activity study of ABC transporter proteins.Results: The resistance index of the MEC/5-FU cells to 5-FU was 36.11.And these cells also showed remarkable resistance to multiple antitumor drugs.Compared to the MEC,the expressions of ABC transporter genes and MDR-1 protein increased in the MEC/5-FU cells.Rh123 accumulated in MEC/5-FU cells was far less than in MEC cells.Conclusion: A multidrug resistance cell line(MEC/5-FU) of human mucoepidermoid carcinoma has been established,which will be useful in studying the drug resistance mechanisms and in screening new drugs for activity,especially against drug resistant tumors.
