摘  要:目的:探讨激活大麻受体对肝癌细胞增殖和凋亡的作用,并对其机制进行初步探讨。方法:将细胞分成对照组、不同剂量大麻受体激动剂WIN-55,212-2(WIN)处理组。MTT法测定WIN对HepG2细胞增殖的影响;DNA梯度电泳法检测HepG2细胞凋亡DNA片段,流式细胞术检测分析各组细胞凋亡率变化,Western blot分析各组细胞caspase-3和c-myc的蛋白表达,分光光度法检测caspase-3、8、9的活性。结果:大麻受体激动剂WIN能抑制肝癌细胞生长和c-myc的蛋白表达,诱导肝癌细胞凋亡,上述效应具有剂量依赖性。而且WIN可能通过诱导细胞caspase-3的蛋白表达和激活caspase-3、8、9活性进而诱导肝癌细胞凋亡。结论:大麻受体激动剂WIN能抑制肝癌细胞生长,诱导肝癌细胞凋亡,并与caspases和c-myc的蛋白表达相关。AIM:To study the effect of the activation of cannabinoid receptor on the proliferation and apoptosis of hepatoma HepG2 cells. METHODS:HepG2 cells were treated with different doses of the activator of cannabinoid receptor-WIN-55,212-2(WIN),MTT was used to detect the effects of WIN on HepG2 cells; Flow cytometry (FCM) was used to examine the effects of WIN on the apop tosis of HepG2 cells. Caspase 3 and c-myc proteins were determined by Western blot assay,and caspase 3,8,9 activity was measured using a colorimetric method. RESULTS:WIN could inhibit HepG2 cell proliferation and induce cell apoptosis dose-dependently. WIN increased caspase 3 protein levels and caspase 3,8,9 activity. CONCLUSION:WIN can induce apoptosis,the effect might be associated with up-regulating expression of caspases protein and down-regulating expression of c-myc protein.
