Study on relationship between neurofilament proteins and Parkinson disease

摘  要:目的研究神经丝蛋白(NFs)作为帕金森病(PD)外周生物标记物的可能性及临床意义。方法用酶联免疫吸附方法检测23例PD患者、30例急性脑梗死患者和29名健康对照者血清NFs水平并进行比较。分析PD患者病程和血清NFs水平的相关性。结果 PD患者和急性脑梗死患者血清NFs水平均明显高于健康对照(P<0.05),而PD患者和急性脑梗死患者之间血清NFs水平差异无统计学意义(P>0.05)。PD患者血清NFs水平和病程具有相关性(r=0.739,P<0.05)。结论 PD患者疾病过程中存在轴索损伤,且轴索损伤严重程度可能随病程延长持续加重;PD患者血清NFs水平改变无疾病特异性,不能作为理想的PD外周生物标记物。Objective To determine if neurofilament proteins could he useful as biomarker for Parkinson disease (PD), and its clinical significanees. Methods Twenty-three PD patients, 30 acute cerebral infarction (ACI) patients, and 29 normal control subjects were recruited in this study. The three groups were matched with age and gender. The level of neurofilament proteins in serum was determined by using a sandwich ELISA. Relation of the course of disease and the level of neurofilament proteins in serum was studied among patients with PD. Results The serum levels of neurofilament proteins were significantly higher in PD and ACI groups than that in normal control group (P〈0.05), but there was no significant difference between PD group and ACI group (P〉0.05). The serum level of neurofilament proteins in PD group had significant correlation with disease duration (r= 0. 739, P〈 0. 05). Conclusions The elevation of serum level of neurofilament proteins in PD patients indicated that degeneration of axons existed in the course of the disease. The severity of axons degeneration might dependent on the length of the disease course. Neurofilament proteins were also significantly higher in ACI group which indicated that it had no disease specificity. Neurofilament protein was not a special biomarker for PD.
