摘 要:目的探讨高压氧(HBO)暴露对Lewis肺癌小鼠抗肿瘤免疫功能的影响。方法90只患Lewis肺癌的C57小鼠被随机分为常压空气组(NA)、高压常氧氮(N2-O2)组和HBO组。NA组置于舱内常压空气下,N2-O2组和HBO组则分别暴露在含10%氧的高压常氧氮和医用纯氧环境下,暴露的压力时程为绝对压250kPa-90分钟,每日1次,每周暴露6天,共暴露两周;加压速度为100kPa/min,减压采用匀速减压法,以10分钟减至常压。实验中检测了各组小鼠的自然杀伤细胞(NK)、非特异杀肿瘤细胞(LAK)、细胞毒性T淋巴细胞(CTL)水平和巨噬细胞(Mφ)杀伤活性;同时还对各组小鼠荷瘤的体积、溃破率及动物致死率进行了观察。结果与NA组相比,HBO组小鼠体内NK、LAK、CTL及Mφ活性显著升高(P<0.01);实验动物肿瘤体积显著减小,荷瘤溃破率则明显增高(P<0.01);且实验组小鼠的存活时间显著延长(P<0.05)。结论HBO暴露显著提高了Lewis肺癌小鼠体内NK、LAK、CTL的活性和增强Mφ细胞杀伤效率。Objective To investigate the effect of hyperbaric oxygen(HBO) on antitumor immune competence in mice with Lewis lung cancer.Methods Ninety C 57 mice with Lewis lung cancer were randomly divided into 3 groups: namely the normal air(NA) group, the normoxic hyperbaric nitrox(N 2 O 2) group and the HBO group. The NA group animals were put into the chamber with normal air pressure. The animals of (N 2 O 2) and HBO groups were exposed to N 2 O 2 mixture gas containing 10% oxygen and therapeutic pure oxygen respectively inside the hyperbaric chamber. The exposure pressure duration was 250kPa(abs) 90min once a day and six times a week for a succession of two weeks. The compression rate was all 100kPa/min, and the decompression was an even 10min period return to normal pressure. The activities of the natural killer(NK), the lymphokine activated killer(LAK), the cytotoxic lymphocyte(CTL) and the abdominal cavity macrophages(M φ) were measured respectively in each group. At the same time, the volume and ulcerative break rate of tumor as well as the fatality of mice with the cancer were observed.Results The activities of NK, CTL and M φ of mice in the HBO group were significantly higher than those in the NA control group( P <0 01), besides the tumor volume was obviously smaller, the ulcerative break rate of tumor was obviously higher and the survival time of mice was also longer( P <0 05).Conclusions HBO exposure could raise the activities of NK, LAK and CTL and also strengthen the killing rate of M φ in mice with the cancer, and these are part of the reasons why the growth of tumor could be inhibited.
