摘 要:目的:建立稳定的、可重复性的局灶性脑缺血再灌注动物模型。方法:采用插线法建立小鼠暂时性大脑中动脉栓塞模型。栓塞时间分别为30、60、90和120min。手术后24h分别对各组小鼠进行神经功能评分。完整端脑2mm冠状厚片行2%红四氮唑(TTC)染色,计算梗死体积。结果:栓塞120min组神经功能评分最高,且变化最小。TTC染色显示,栓塞120min可引起该侧大脑中动脉供血区域广泛的梗死灶,而且梗死灶大小适宜,稳定性好。结论:插线法是一种简单、可靠的制作小鼠暂时性大脑中动脉栓塞模型的方法,栓塞120min再灌注22h可引起稳定的梗死灶,适合于脑缺再灌注损伤的对照研究。Objective:To establish stable and repeatable focal cerebral ischemia reperfusion model in mice.Methods:Male,ripe Balb/C mice were selected to establish the instant middle cerebral artery occlusion(MCAO)model by intraluminal occlusion using monofilament.The times of MCAO were 30,60,90 and 120 min respectively.24 h after operation,nervous function grades of mice were carried out.TTC staining of 2 mm brain slice was used to calculate the infarct volumes.Results:Statistic analysis showed that nervous function grade of the group of MCAO 120 min /reperfusion 22 h is highest and most stable.TTC staining showed that MCAO 120 min /reperfusion 22 h could induce broad infarct areas which obtained blood supply from MCA,the volumes of infarct areas are stable and fit for experiment researches.Conclusion:Intraluminal occlusion of MCA using monofilament is a kind of simple and reliable method for mouse instant MCAO model.And MCAO 2 h /reperfusion 22 h can induce stable infarct areas,which is fit for the control studies of cerebral ischemia reperfusion.
