摘 要:目的建立同时测定血浆中两种不同形式维生素B6(吡哆醇Pyridoxine,PN和吡哆醛Pyridoxal,PL)的高效液相色谱法。方法采用Luna C18色谱柱(150 mm×4.6 mm,5μm),外标法,荧光检测器(波长为λE326 nm,λM400 nm),流动相:甲醇-磷酸缓冲液(12∶88),pH3.2,流速:1.2 ml/min进行分析。结果在2.5~160μg/ml浓度范围内,血中PN、PL测定的平均回收率分别为(86.0±4.74)%和(96.1±3.52)%,RSD分别为4.25%和3.85%。血中PN、PL的最低检出浓度分别是2.37、1.28μg/ml。结论该法灵敏、快速、准确、重复性好。Objective To establish a High Pressure Liquid Chromatography method by which different types of Vitamin B6(such as Pyridoxine,PN and Pyridoxal,PL) in blood could be determined simultaneously.Methods in the outside-standardizing method,samples were determined using Luna C18 analysis column(150 mm×4.6 mm,5 μm) and fluorescence detector(λE 326 nm;λM 400 nm) with a mobile phase consisting of 35mmol/L sodium phosphate containing 2.5mmol/L heptanesulfonic acid,adjusted to pH 3.2 with 85% orthophosphoric acid and 12% methanol(V∶V) at 1.2ml/min.Results In the concentration range of 2.5 to 160 μg/ml,the average recycle rates of PN and PL in blood were(84.62±3.603)% and(95.66±3.695)%.The variation index of repeated determinations was 4.25% and 3.85% respectively.The detectable limit inferior concentration of PN and PL in blood was 2.37 and 1.28 μg/ml respectively.Conclusion A determination method of PN and PL by HPLC was constructed,being sensitive,accurate,quick and reproducible.
