bFGF与阿朴吗啡对形觉剥夺性近视抑制效果的比较研究

摘　　要：目的:通过药物实验比较bFGF与阿朴吗啡对形觉剥夺性近视的抑制效果,探讨此两种药物抑制近视的作用机理。方法:半透明的塑料眼罩遮盖法建立72只出生2d的健康艾维因雏鸡FDM模型,随机分为8组:正常对照组8只(A),FDM组16只(B),FDM+bFGF200ng 8只(C),FDM+bFGF500ng 8只(D),FDM+bFGF1000ng 8只(E);FDM+阿朴吗啡200ng 8只(F),FDM+阿朴吗啡500ng 8只(G),FDM+阿朴吗啡1000ng 8只(H)。单眼遮盖1w后所有实验动物检影验光,取A组及随机取B组8只鸡给予腹腔麻醉后处死,取出眼球,游标卡尺测眼轴,并取后极部固定备用。C、D、E、F、G、H,6组单眼遮盖1w后,右眼行结膜下注射1w后检影验光,游标卡尺测眼轴,取后极部观察其形态学变化。结果:鸡形觉剥夺1w后近视形成,眼轴延长。药物治疗1w后C、D、E、F、G、H各组与B组右眼屈光度眼轴前后径及赤道径,两两比较差异均有统计学意义;C、D、E各组间两两比较C组与D、E两组差异均有统计学意义;F、G、H各组间眼轴与屈光度两两比较差异均有统计学意义。近视鸡视网膜各层变薄,光镜下可见病理性改变。结论:bFGF与阿朴吗啡均能明显抑制形觉剥夺性近视的发展,阿朴吗啡对形觉剥夺性近视的抑制效果呈剂量相关性,相比之下bFGF对形觉剥夺性近视的抑制效果更佳。Objective： Compare the inhibition effects of basic fibroblast growth factor（bFGF） and apomorphine to From-deprivation myopia by medicine test and explore the path-ological mechanism how the both medicines inhibit myopia.Methods：we established 72 2-day old baby chickens FDM models by semitrans-parent plastic valance cover.we randomly divided these chickens into 8 groups,except group B 16 members each of which 8 members： normal controls,Group A.FDM group,Group B.FDM,with bFGF 200ng injection,Group C.FDM with bFGF 500ng injection,Group D.FDM,with bFGF 1 000ng injection,Group E.FDM,with apomorphine 200ng injection,Group F.FDM,with apomorphine 500ng injection,Group G.FDM,with apomorphine 1000ng injection,Group H.One week after covered all the chickens were examined the eyesight.Group A and eight of group B,d axiallength was measured.Then keep the tissue of retina and sclera.For Group C,D,E,F,G,H,eyesight was examined and axial length was measured one week after injection.Then morphological changes of retina and sclera tissue in the posterior pole were examined.Results：Myopia was formed and axis oculi was extended after from-deprivation.We compared the difference of anteroposterior diameter and equatorial diameter between each of the last 6 groups and Group B one week after injection,we found the difference was significant.There were significant differences between Group C and D,Group C and E,Group F and G,Group F and H,Group G and H.Each layer of retina was thinningz in the light macroscope.Conclusion：Both bFGF and apomorphine could dose dependently inhibit the progression of FDM.The inhibit of apomorphine is dose-dependent.By comparison,bFGF was a more strong inhibitor.
