摘 要:目的研究N-乙酰氨基葡萄糖对体外培养的小鼠B16黑素瘤细胞增殖和凋亡、酪氨酸酶活性及黑素生成的影响。方法采用MTT法、流式细胞仪技术和投射电镜方法测定N-乙酰氨基葡萄糖对培养的小鼠B16黑素瘤细胞的增殖抑制作用和细胞的凋亡,体外氧化DOPA反应方法测定酪氨酸酶活性,NaOH法测定黑素生成量。结果 25mM、50mM和100mMN-乙酰氨基葡萄糖明显抑制B16黑素瘤细胞增殖、诱导细胞凋亡并抑制黑素生成,50mM和100mM N-乙酰氨基葡萄糖明显抑制酪氨酸酶活性,差异具有统计学意义(P<0.05或P<0.01)。结论 N-乙酰氨基葡萄糖抑制培养的小鼠B16黑素瘤细胞的黑素生成,可能具有开发成为美白产品的前景。Objective To study the effects of N-acetylglucosamine on cell proliferation, cell apoptosis, tyrosinase activity and melanogenesis in Murine B 16 melanoma cells. Methods Effects of N-acetylglucosamine on cell proliferation, apoptosis, tyrosinase activity and melanogenesis in cultured Murine B 16 melanoma cells were measured by MTT method, flow cytometry, transmission electron microscope, oxidative DOPA reaction and NaOH respectively. Results Cell proliferation and melanogenesis were inhibited and cell apoptosis was induced by 25mM, 50mM and 100mM N-acetylglucosamine, tyrosinase activity was inhibited by 50mM and 100mM N-acetylglucosamine in B 16 melanoma ceils significantly (P〈0.05 or P〈0.01). Conclusions Vitro studies show that N-acetylglucosamine inhibits melanogenesis, and thus may be prospectively developed to be a skin lightening agent.
