TGF-β_1对绒癌JEG-3细胞MMP-9 mRNA表达的影响

摘　　要：目的:研究外源性TGF-β1作用不同时间对绒癌JEG-3细胞基质金属蛋白酶-9(MMP-9)mRNA表达的影响。方法:研究对象为永生化绒癌JEG-3细胞系,以100ng/ml浓度的TGF-β1分别作用JEG-3细胞0h、12h、24h、48h和72h,采用RT-PCR法检测各组细胞MMP-9 mRNA的表达。结果:随着TGF-β1作用时间的延长,MMP-9 mRNA的表达逐渐增加(P<0.01)。结论:绒癌JEG-3细胞MMP-9 mRNA的表达对外源性TGF-β1的刺激呈现一定的时间效应,可能与绒癌的侵袭转移相关。Objective：To study the effects of ectogenic TGF-β1 with different action time on MMP-9 mRNA expression of choriocarcinoma JEG-3 cells.Methods：The MMP-9 mRNA expression of choriocarcinoma JEG-3 cells was detected by RT-PCR after the JEG-3 cells were stimulated by TGF-β1 for 0h,12h,24h,48h and 72h respectively.Results：Along the action time of TGF-β1,the MMP-9 mRNA expression of choriocarcinoma JEG-3 cells increased gradually（P〈0.01）.Conclusions：The MMP-9 mRNA expression of choriocarcinoma JEG-3 cells appears time effect to stimulation of ectogenic TGF-β1,which may be concerned with invasion and metastasis of choriocarcinoma.
