摘 要:目的比较HE染色和酶组织化学染色法评价高强度聚焦超声(HIFU)消融兔乳腺移植瘤效果的准确性。方法 18只兔乳腺移植瘤模型,随机分成对照组6只、消融组12只,消融组再分成A、B组各6只分别用80 W、160 WHIFU于肿瘤中央定点消融5 s。消融后即刻取标本行HE染色、琥珀酸脱氢酶(suecinate dehy drogenase,SDH)酶组织化学染色光镜下观察。结果 A、B两组HIFU消融后HE染色显示瘤细胞形态无明显改变,酶组织化学染色显示A组靶区SDH活性散在阳性,提示还残留有活性的瘤细胞,B组SDH活性彻底消失,提示瘤细胞已经死亡,两组比较差异显著(P<0.05)。结论酶组织化学法染色测定SDH活性比HE染色能更客观准确地评价HIFU消融兔乳腺移植瘤的效果。Objective To campare the accuracy of HE staining and enzyme histochemical staining to estimate efficacy of high intensity focused ultrasound (HIFU) in ablation of implanted raBbit Breast tumor. Methods 18 implanted rabbit breast tumormodels were divided into control group (n=6) and HIFU ablation group (n=12) and HIFU ablation group was redivided into A (n=6) and B (n=6) ablated in fixed point within tumor's centre by HIFU at 80 W and 160 W for 5 seconds respectively. After microwave ablation, paraffin sections and frozen sections of the tumors were prepared for routine HE staining and enzyme histochemical staining with succinate dehydrogenase (SDH), and observed under light microscope, Results Shortly after HIFU ablation, the morphology and arrangements of the nucleus of the ablated tumor cells in A and B group showed no obvious alteration in HE stained sections, but in sections with enzyme histochemical staining, sporadic activity of the enzyme was detected in A group's coagulated tumor at 80 W, indicating remnant tumor cells surviving. The activity of SDH in B group's ablated tumor tissue at 160 W disappeared, suggesting the death of tumor cells. The ablation effect was markedly different between the two groups (P〈0.05). Conclusions SDH staining is more accurate and more objective than HE staining in assessing effect of HIFU ablation in implanted rabbit breast tumor.
