牙龈卟啉单胞菌表面相关物质通过Fas-FasL途径诱导T淋巴细胞凋亡的体外研究

摘  要:目的检测Fas/FasL在牙龈卟啉菌表面相关物质(SAM)诱导的T淋巴细胞凋亡中的表达。方法选取10名全身及牙周组织健康受试者,分离外周血中单核细胞(PBMC),用SAM体外刺激PBMC,用特殊的单克隆抗体(Fas、FasL)标记,并进行流式细胞仪检测。结果 FasFasL的表达量明显上调。实验组Fas表达量在24h为25.38%±3.17,在96h为65.11%±3.58;实验组FasL的表达量在24h为5.23%±1.53,在96h为20.59%±2.68。用抗Fas单克隆抗体阻滞Fas-FasL相互作用导致T细胞凋亡的明显减少,百分比为20.53%±3.26,未加抗体的为48.47%±4.15。但残余的细胞凋亡活动与阴性对照相比仍高。结论 SAM诱导T淋巴细胞凋亡主要通过Fas-FasL途径,并具有时间依赖性,通过数据证明了细胞凋亡的分子机制。Objective To investigate the involvement of the Fas/FasL-mediated apoptotic pathway in surface-associated material from Porphyromonas gingivalis (SAM) -induced T-cell apoptosis.Methods Ten systemically and periodontally healthy subjects participated in this study. Venous blood was drawn and peripheral blood mononuclear cells (PBMC) was isolated. T lymphocytes stimulated with SAM and the cells were labled by the special monoclonal antibodies and analyzed by flow cytometry.Results The stimulation of T cells with SAM leads to the up-regulation of Fas and FasL. Fas:25.38%±3.17 at 24h,65.11%±3.58 at 96h; FasL:5.23%±1.53 at 24h,20.59%±2.68 at 96h. The blocking of Fas-FasL interaction with anti-Fas monoclonal antibody led to significantly reduce T-cell apoptosis of 20.53%±3.26,while without anti-Fas antibody led to T-cell apoptosis of 48.47%±4.15,but the residual apoptosis activity was higher than that of background remained.Conclusion SAM induces apoptosis of T lymphocytes by the Fas-FasL pathway. Data obtained on the molecular mechanism of apoptosis and the effect was time-dependent.
