摘  要:目的探讨3,3-二吲哚基甲烷(DIM)联合肿瘤坏死因子相关凋亡诱导配体(TRAIL)对前列腺癌细胞的抑制作用。方法CCK-8法测定DIM、TRAIL及DIM联合TRAIL作用于PC-3细胞后的细胞生存率,同时用流式细胞术测定细胞凋亡率,Western印迹检测caspase-3表达水平的变化。结果各给药组与对照组比较均能不同程度抑制PC-3细胞增殖并诱导其凋亡,且caspase-3表达上调;DIM与TRAIL联用明显增强诱导凋亡作用。结论DIM与TRAIL联用均可明显增强PC-3细胞抑制效果和诱导凋亡作用,其凋亡作用机制可能与其上调caspase-3表达有关。Objective To investigate the inhibitory effect of 3,3-diindolylmethane (DIM) in combination with TNF-related apoptosis-inducing ligand (TRAIL) on the prostate cancer cells. Methods CCK-8 methods was used for detecting the survival rate of PC-3 cells treated with DIM,TRAIL,or DIM-TRAIL. The apoptosis rate of PC-3 cells was tested by flow cytometry (FCM). The expression of caspase-3 was measured by Western blot. Results Compared with that of control group,the proliferation of PC-3 cells was inhibited and the apoptosis was induced differently in all treatment groups,and the expression of caspase-3 was up-regulated. The combined use of DIM-TRAIL induced apoptosis of PC-3 cells. Conclusions Combination use of DIM and TRAIL could obviously enhance the inhibitory effects and apoptosis rate on PC-3 cells. The possible mechanism may be the up-regulation of expression of caspase-3.
