摘  要:目的:检测人牙周膜成纤维细胞(human periodontal ligament fibroblasts,hPDLF)中胆碱乙酰转移酶(choline acetyltransferase,ChAT)的表达,并观察尼古丁对其表达的影响,以探讨ChAT在吸烟与牙周炎发生发展过程中的可能作用。方法:体外培养hPDLF,免疫组化和RT-PCR方法检测hPDLF中ChAT表达,并观察尼古丁(10~12mol/L)作用于hPDLF24h后ChAT表达的变化。结果:免疫组化染色hPDLF胞浆中ChAT表达阳性;RT-PCR检测hPDLF中ChATmRNA阳性表达。尼古丁作用后ChAT蛋白和mRNA表达均明显上调与对照组相比差异有统计学意义(P<0.05)。结论:hPDLF中存在非神经元型胆碱能系统物质ChAT的表达,尼古丁可上调其表达。AIM:To examine whether human periodontal ligament fibroblasts (hPDLF) express Choline acetyltransferase (ChAT),a non-neuronal acetylcholine system substance,and to observe the effects of nicotine on of ChAT in hPDLF.METHODS:Expression of ChAT in hPDLF was detected by immunohistochemistry (IHC) at protein level and by RT-PCR at mRNA level.The effects of nicotine on ChAT was observed by treating hPDLF with nicotine (10-12mol/L) for 24 h.RESULTS:Positive staining of ChAT was detected by IHC in the cytoplasm of hPDLF.mRNA expression of ChAT in hPDLF was confirmed by RT-PCR.Nicotine treatment significantly increased both the mRNA and protein expression of ChAT in hPDLF (P0.05).CONCLUSION:hPDLF expresses ChAT and nicotine can up-regulate ChAT expression at mRNA and protein levels in hPDLF.
