摘  要:目的:已采用功能性抗体库技术筛选出抗肺癌的单抗13C9,进一步确定其识别的抗原在肺癌组织和细胞中的表达及体内抑制肺癌生长的能力,以期为肺癌靶向治疗提供有价值的分子靶位和治疗剂。方法:常规方法鉴定13C9单抗的一般特性,用细胞免疫荧光、免疫组织化学检测其抗原蛋白在肺癌细胞及组织中的表达和定位,体内移植瘤治疗实验评价13C9单抗对肺癌生长的抑制作用,Western blotting等方法鉴定13C9单抗识别的抗原。结果:13C9单抗与肺癌细胞GLC-82和A549的细胞膜结合,免疫组化结果显示其识别的抗原在人肺癌组织中高表达(79.8%),而在癌旁组织中不表达或低表达(9.6%,P<0.05)。该抗体亚型为IgM、κ型,13C9单抗治疗能够抑制人肺腺癌移植瘤的生长,抑制率达63.29%(P<0.05)。Western blotting显示13C9单抗识别的抗原相对分子质量约220 KD。结论:单抗13C9能显著抑制体内肺癌生长,具有成为肺癌靶向治疗剂的应用潜力,其识别的特异表达在肺癌细胞膜上的220 KD的膜蛋白,可能是肺癌潜在的治疗靶位。Objective:Functional monoclonal antibody library had been adopted to screen for monoclonal antibody 13C9 suppressing lung cancer.We recognize the expression of the specific antigen on 13C9 in lung cancer and the ability to inhibit the growth of lung cancer in vivo to provide valuable molecular targets and treatment agents for targeted therapy of lung cancer.Methods:Conventional methods were used to identify the general characteristics of 13C9 monoclonal antibody.The 13C9 antibody was studied by immunofluorescence,immunohistochemistry to detect its antigen protein in lung cancer cells and tissues and the antigen protein's localization.Tumor treatment of the 13C9 antibody in vivo was tested to evaluate the growth inhibition of lung cancer.The corresponding antigen of 13C9 was identified by western blotting.Results:The 13C9 antibody could recognize cell surface proteins of lung cancer cell lines GLC-82 and A549,also highly recognize the antigen in lung cancer tissue by 79.8%,weekly or barely reacted with normal lung tissue by 9.6%(P〈0.05) reversely.The 13C9 antibody identified as IgM,Kapa could inhibit lung tumor growth in mice by 63.29%(P〈0.05).Western blotting revealed that the molecular weight of the antigen of 13C9 antibody was about 220 KD.Conclusion:The 13C9 antibody can significantly inhibit the growth of lung cancer in vivo,has become a potential targeting therapeutic agent for lung cancer,the membrane protein about 220 KD on lung cancer specific identified by the 13C9 antibody may be therapeutic target site of lung cancer.
