摘 要:目的 观察芹菜素对大鼠脑缺血不同时间再灌注损伤后丙二醛(MDA)、超氧化物歧化酶(SOD)表达的影响,探讨芹菜素的神经保护作用,为脑缺血再灌注损伤的防治提供新药可能.方法 将大鼠72只随机分为假手术组(S组)、模型组(M组)及芹菜素组(A组),后2组按再灌注时间不同又分为再灌注24、48、72h和7d各4亚组,总共9小组.每组8只大鼠.采用线栓法制备大鼠左侧大脑中动脉缺血(1.5h)再灌注模型.造模后按Zea Iorlga法评定神经功能缺损状况.观察至规定时间断头取脑,制备脑组织匀浆,用黄嘌呤氧化酶法和硫代巴比妥酸法分别测定脑组织SOD活性和MDA含量.结果 与S组比较,M组在脑缺血再灌注后各时间点,MDA含量均明显升高(P〈0.05或0.01),SOD活性均明显下降(P〈0.05或0.01);M72h组MDA含量明显高于M24h和M7d组,差异有统计学意义(P〈0.05).芹菜素干预后,脑组织MDA含量在脑缺血再灌注后48h和72h有所降低(P〈0.05),而SOD活性在缺血再灌注后48、72h和7d有所增高(P〈0.05).大鼠脑缺血再灌注后均出现神经功能缺损症状,M组在脑缺血再灌注7d神经行为学评分较之前各时间点均有明显改善,芹菜素在脑缺血再灌注后72h有改善神经功能的作用.大鼠脑缺血再灌注后24h和7d,神经行为学评分与MDA含量呈正相关关系(r=0.516,P=0.041;r=0.582,P=0.018);与SOD活性呈负相关关系(r=-0.565,P=0.023;r=0.599,P=0.014).结论 芹菜素对脑缺血再灌注后自由基损伤有一定保护作用.Objective To investigate the neuroprotetive effect of apigenin on cerebral ischemia-reperfusion (I/R) in rats. Methods Seventy two rats were randomly divided into three groups: model group (M), sham operation group (S) and apigenin group (A), the cerebral I/R model were established by middle cerebral artery occlusion (MCAO). The brain levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were determined with xanthine oxidase method and thiobarbiturate at 24h, 48h, 72h and 7d after I/R, respectively. Results Compared to group S, the levels of MDA in brain tissue of group M were increased and the activities of SOD decreased at all time points after reperfusion (P〈0.05 or P〈0.01). MDA content in brain of group M72h increased markedly compared to that in group M24h and group M7d (P〈0.05). After apigenin treatment, the contents of MDA were decreased at 48h and 72h after reperfusion (P〈0.05), and the activities of SOD were increased at 48h,72h and 7d after reperfusion (P〈0.05), compared to the same time points of group M. Conclusion Apigenin can protect cerebral tissue from I/R injury through inhibiting effects of free radicals.
