以CK20为标志物对乳腺癌微转移检测的研究

摘　　要：目的探讨乳腺癌微转移的标志基因。方法采用逆转录聚合酶链反应(RT-PCR)方法 ,检测了20例乳癌病人正常乳腺组织、癌组织、淋巴结、外周血、10例乳腺纤维瘤,10例非肿瘤病人淋巴结及外周血中细胞角蛋白19(CK19)、角蛋白20(CK20)的表达。结果乳癌组织、正常乳腺组织、转移淋巴结、纤维腺瘤中CK19、CK20均有很高的表达率。CK20在非肿瘤病人淋巴结和外周血中无表达,而CK19有40%表达。结论来源于上皮细胞的良、恶性肿瘤均遗留上皮细胞表达角蛋白的特征;CK20可以作为乳腺癌病人病理检查为非转移淋巴结和外周血中微小转移的标志基因,CK19则不适合。Objective To study the micro-metastasis of breast cancer by marker genes.Methods The detection of CK19 and CK20 expression by using RT-PCR in 20 cases of breast cancer.Normal breast tissue,cancer tissue,lymph nodes and peripheral blood samples of 20 cases of breast cancer and 10 cases of breast fibromatosis,lymph nodes and peripheral blood samples of non-cancerous patients were examined.Results The rates of expression of CK19 and CK20 in breast cancer tissue,normal breast tissue,metastatic lymph nodes and fibroadenoma tissue were very high.CK20 didn＇t express in lymph nodes and peripheral blood samples of non-cancerous patients,and CK19 was expressed in 40% of them.Conclusion Benign and malignant tumors derived from epithelial cells were inherited with keratin （CK） features.CK20 marker gene can be used as a non-pathological examination of breast cancer patients with metastatic lymph nodes and micro-metastasis in peripheral blood,and CK19 is not suitable for this purpose.
