摘　　要：目的分析B细胞非霍奇金淋巴瘤(B-NHL)患者外周血中T细胞信号转导分子TCRζ基因的表达特点。方法采用SYBRGreenⅠ实时定量RT-PCR和相对定量分析法检测17例B-NHL患者外周血单个核细胞(PBMCs)中TCRζ基因的表达情况,以β2微球蛋白基因(β2M)作为内参,采用相对定量公式:2-△Ct×100%,计算TCRζ基因的相对mRNA表达量,17例健康成人作为对照。结果全部B-NHL样本均全部表达TCRζ基因,但其表达水平显著低于对照组(P=0.000),健康人PBMCs的TCRζ基因mRNA的表达水平为B-NHL患者的3.26倍。B-NHL患者TCRζ基因表达水平与患者的年龄无相关性。结论 B-NHL患者中TCRζ基因的mRNA表达水平明显下调,可能是其细胞免疫功能缺陷的原因之一。Objective To analyze the expression level of signaling transduction factor TCR ζ chain gene in T cells from patients with B-cell non-Hodgkin＇s lymphomas（B-NHL）.Methods Real-time RT-PCR with SYBR GreenⅠtechnique was used for detecting the mRNA level of TCR ζ chain gene in peripheral blood mononuclear cells（PBMCs）of 17 patients with B-NHL,and 17 cases healthy adults served as control.The β2-microglobulin gene（β2M）was used as an endogenous reference.Relative mRNA expression level of TCR ζ chain gene was analyzed by the 2-△Ct×100% method.Results TCR ζ chain gene was expressed in all patients with B-NHL.Compared with healthy control,the relative mRNA level of TCR ζ chain gene of patient was significantly decreased（P=0.000）.All samples from healthy individuals expressed normal level of TCR ζ chain gene,which is 3.26 times higher than that from B-NHL patients.There was no significant correlation between the expression level of TCR ζ chain gene and age of patients with B-NHL.Conclusion The low mRNA level of TCR ζ chain gene could be identified in peripheral blood of patients with B-NHL,which might related to cellular immunodeficiency in most of B-NHL patients.
