摘  要:用免疫组织化学方法研究RCS大鼠(Royal College of Surgeon's rat,RCS rat)视网膜变性过程中早、中、晚期二级神经元形态的变化。结果发现,P1MRCS大鼠各二级神经元未见明显的形态改变。P2M RCS大鼠视网膜外核层萎缩约85%,视杆双极细胞顶端树突萎缩;水平细胞的树突与轴突在外网状层的分层未见改变。P3M RCS大鼠视网膜外核层萎缩近95%,RCS大鼠视杆双极细胞顶端萌生新的神经突起;水平细胞的树突分支明显丢失,轴突在外网状层的分层发生改变,出现新生神经突起;无长突细胞的树突在内网状层的分层至变性晚期也未见改变。该研究表明RCS大鼠视网膜二级神经元形态的改变是继发性改变,是感光细胞变性后对传入缺失的一种反应,即重构反应。在进行视网膜功能救治时需要考虑重构反应带来的影响。This study examined the morphologcal modification of the second-order neurons during retinal degeneration in RCS rats.The RCS-p+ rats and control rats were sampled at P1M,P2M,P3M three groups,n=3 in each group.The animals were anesthetized and retinas were dissected following standard protocol.Retinal cryosections were immunostained with PKC-α,calbindin,calretinin and neurofilament antibody,and were photographed using an Olympus confocal microscope(FV500) to examine the morphologcal changes of the second-order neurons during retinal degeneration.Compared with the control rats,it was found that there was no observable changes in the second-order neurons in P1M RCS rats.Rod bipolar cells ' dendrites disappeared in P2M RCS rats,but many new neurites sprouted from rod bipolar cells in P3M RCS rats.The dendrites of horizontal cells reduced significantly,the horizontal cells' axon sprout neurites extending into the outer nuclear layer in P3M RCS rats.The lamination of amacrine cells ' processes in IPL was normal in P3M RCS rats.These results suggest that morphologcal modification of the second-order neurons in RCS rats is resulted from photoreceptor apoptosis and the deprivation of inputs.The maintenance of stable structure of the second-order neurons in RCS rats depended on function of photoreceptors.Remodeling may stimulate the transplanted cells or tissues to form new synaptic,or form the abnormal neurocircuitry to affect the transmission of visual information.Therefore,when considering therapeutic programs to reconstruction the retina's function,we need to consider the impact of remodeling to treatment.
