PTEN、COX-2及MMP-9在骨肉瘤中的表达及其意义

摘　　要：[目的]探讨张力蛋白同源（PTEN）、环氧化酶-2（ COX-2）及基质金属蛋白酶9（MMP-9）在骨肉瘤中的表达及其意义.[方法]选取骨肉瘤石蜡包埋标本28例,以25例骨软骨瘤石蜡包埋标本作为对照.采用SP法对以上病理标本进行免疫组化染色.检测PTEN、COX-2及MMP-9的表达情况.[结果]PTEN在骨肉瘤组织中的表达率为57.1%（16/28）,显著低于其在骨软骨瘤组织中的表达率为88.0%（22/25）,两者的差异有显著性（P〈0.05）;COX-2和MMP-9在骨肉瘤中的表达率78.6%（22/28）及82.1%（23/28）,显著高于在骨软骨瘤中的表达率分别为36.0%（9/25）和28.0%（7/25）,差异有显著性（P〈0.05）.PTEN在不同分期以及不同病理亚型骨肉瘤组织中的阳性表达差异无显著性（P〉0.05）.COX-2及MMP-9表达与病理亚型无明显相关,而与临床分期关系密切.PTEN与COX-2和MMP-9的表达具有负相关性;COX-2与MMP-9的表达具有正相关性.[结论]PTEN、COX-2及MMP-9与骨肉瘤的发生、发展有相关性,联合检测对于骨肉瘤的诊断和预后判断具有一定的意义.[Objective] To investigate the expression of PTEN, COX-2 and MMP-9 in osteosarcoma and their relationship with local invasion. [Methods] Paraffin imbedding specimens of 28 cases of osteosareoma and 25 eases of osteoenchondroma were chosen to study the expression of PTEN, COX-2 and MMP-9 with SP immunohistochemistry method. [Results]The expression rate of PTEN, COX-2 and MMP-9 in osteosarcoma tissues were 57. 1% （16/28）, 78. 6% （22/28）and 82. 1%（23/28）, respectively. The expression of PTEN in osteosarcoma was lower than that in osteoenchondroma（ P 〈0.05）, but that of COX-2 and MMP-9 in osteosarcoma were higher than that in osteoenchondroma（ P〈0.05）. The positive expression of PTEN in different stages or in different pathological subtypes had no significant difference（ P〈0.05）. The expression of COX-2 and MMP-9 also had no significant difference among different pathological subtypes, but had significant difference among different stages（ P 〈0.05）. The expression of PTEN had negative correlation with COX-2 and MMP-9, and there had direct correlation between COX-2 and MMP-9. [Conelusion]PTEN, COX-2 and MMP 9 are correlated with the genesis and development of osteosarcoma. Combination measurement of PTEN, COX-2 and MMP-9 will help to judge the diagnosis and prognosis of osteosarcoma.
