摘 要:[目的]探讨水通道蛋白1(AQP1)在α-萘异硫氰酸(ANIT)诱导的肝内胆汁淤积大鼠肝胆系统的表达情况.[方法]将健康雄性Wistar成年大鼠16只随机分为ANIT组和对照组, 每组8只.ANIT组由10 g/L ANIT花生油溶液一次性灌胃诱导肝损伤.对照组仅灌花生油.检测两组AQP1的表达.平均光密度(OD)值.[结果]两组大鼠肝胆系统均有AQP1表达.对照组OD值为(0.38±0.08)明显高于ANIT组(0.10±0.03)(P〈0.01).[结论]ANIT诱导的胆汁淤积大鼠肝胆系AQP1的蛋白表达下调,提示AQP1参与胆汁淤积的病理机制.[Objective]To investigate the change of aquaporinl(AQP1) expression in hepatobiliary system of rats with alpha-naphthylisothioeyanate (ANIT)-indueed cholestasis. [Methods]Sixteen healthy Wistar male adult rats were randomly divided into ANIT group and control group with 8 for each. The control group was treated with only peanut oil. The ANIT group was treated with ANIT peanut oil. A single oral dose of 150 mg/kg ANIT dissolved in peanut oil was given to rats by gavage(10g/l) to induce liver injury. Itnmunohisto- chemical method was used to detect AQP1 expression. The image of three sections of each rat was analyzed and the average gray level was determined. [Results]A single oral ANIT could induce cholestasis successfully. The expression of AQP1 protein was detected in two groups. AQP1 protein level in ANIT group was 0.10± 0.03 which was significantly lower than that(0.38±0.08) in control group( P 〈0.01). [Conclusion] The AQP1 expression in hepatobiliary system of rats with ANIT-induced cholestasis is significantly decreased. It suggests that AQP1 involves in the pathological mechanism of cholestasis.
