犬瘟热病毒水貂株的分离与鉴定

摘　　要：从疑似犬瘟热病死水貂的肝脏中分离出1株病毒,并进行了系统的鉴定。结果表明:该毒株接种于Vero传代细胞后,出现了典型而有规律的病变;其病毒理化特性与犬瘟热病毒相同;致细胞病变作用可被兔抗CDV阳性血清阻断;间接免疫荧光试验表明接种病毒的BHK-21细胞出现特异性的亮绿色荧光,而正常细胞则未见绿色荧光;对提取接种病料后的Vero细胞培养物中的总RNA进行RT-PCR扩增,得到了324bp和1053bp的目的片段,证明该毒株为CDV。A strain of canine distemper virus（CDV）was isolated and identified systemically from the liver of a dead mink suspected of having canine distemper.Results indicated that typical and disciplinary pathological changes appeared on Vero cells after being inoculated with the treated pathological material,and the physical and chemical characteristics of the isolated virus were similar to those of CDV.The cytopathogenic effect was interdicted by rabbit anti-CDV positive serum.Indirect immunofluorescent assay showed that idiosyncratic bright green fluorescence appeared on BHK-21 cells inoculated with CDV,but not in the normal BHK-21 cells.The total RNA extracted from Vero cell culture inoculated with the virus were amplified by reverse transcription-polymerase chain reaction（RT-PCR）,and two objective segments（324bp and 1053bp）were obtained.Thus,the isolated virus strain was proved to be CDV.
