二白连地汤治疗大鼠溃疡性结肠炎的实验研究

摘　　要：目的:以硫酸葡聚糖5000(DSS)诱导建立大鼠溃疡性结肠炎模型,观察二白连地汤对大鼠溃疡性结肠炎的治疗效果。方法:将48只大鼠随机分为4组:正常组、模型组、柳氮磺吡啶组(SASP)、二白连地汤组。正常组自由饮用蒸馏水,模型组、SASP组及二白连地汤组自由饮用含5%DSS的蒸馏水诱导建立大鼠溃疡性结肠炎模型,7d后正常组及模型组每天给予生理盐水灌胃,而SASP和二白连地汤组分别给予SASP及二白连地汤灌胃治疗,每日观察大鼠体质量和大便性状,并采用疾病活动指数(DAI)对动物状态进行评分,2周后处死所有大鼠,观察其结肠大体形态改变及镜下病理学变化并进行评分。结果:模型组大鼠DAI评分、大体形态及病理学评分明显高于正常组(P<0.01)。SASP组及二白连地汤组DAI评分、大鼠大体形态及病理学评分明显低于模型组(P<0.01),但SASP组与二白连地汤组DAI评分、大鼠大体形态及病理学评分无统计学差异(P>0.05)。结论:DSS可成功诱导建立大鼠溃疡性结肠炎模型;二白连地汤有利于炎症的消除和组织修复,能有效治疗溃疡性结肠炎。Objective:To observe the effect of erbailianditang on ulcerative colitis rats induced by intragastric administration with DSS. Methods:48 Male Wistar rats were randomly divided into four groups:Group B(blank group),Group M(model group),Group S(SASP-treated group),Group E ( erbailianditang- treated group). The model of ulcerative colitis rats was established in Group M,S and E by administration with DSS. Then the rats of Group B and M were administrated with 5ml normal sodium,Group S and E with same dose SASP and erbailianditang respectively for 14 days. The weight,stool viscosity and hematochezia were observed and accumulated as disease active index (DAI) score; After treatment,all rats were killed. HE staining was performed to detect the pathological change in colon of all rats. Result:Compared to Group B,the mark of DAI,body configuration,pathological change in Group M had a significant increase (P<0.01). Compared with Group M,the mark of DAI,body,configuration,pathological change in Group S,E had a significant decrease (P<0.01). Compared with Group S,the mark of DAI,body,configuration,pathological in Group E had no significant difference (P>0.05). Conclusion:Intragastric administration with DSS can successfully induce the model of ulcerative colitis rats. Erbailianditang can help clear inflammation and repair tissue and effectively treat the ulcerative colitis rats.
