摘 要:目的:探讨一种新CD44变异体(CD44v17)在乳腺癌组织中的表达及其临床意义。方法:根据本实验室在MCF-7/Adr中新发现的一种CD44剪切拼接变异体CD44v17(基因库NO.FJ216964)设计特异性引物,应用SYBRG reenⅠ荧光染料,采用实时PCR法检测了52例乳腺癌组织及相应癌旁组织CD44v17 mRNA的表达情况,根据标准曲线计算出CD44v17 mRNA的表达量,分析CD44v17 mRNA表达与乳腺癌临床生物学特性之间的关系。结果:乳腺癌组织CD44v17 mRNA表达(3.10±1.12)明显高于相应癌旁正常组织(2.31±0.84)(P<0.01);乳腺癌淋巴结转移组(4.07±1.13)显著高于无淋巴结转移组(2.75±0.89)(P<0.01);肿瘤组织>5 cm组CD44v17 mRNA表达(4.18±0.95)明显高于肿瘤组织≤5 cm组(2.96±1.07)(P<0.05);髓样癌(5.27±0.30)显著高于导管内癌(3.28±1.08)和浸润性导管癌(2.92±1.04)(P<0.05),导管内癌和浸润性导管癌CD44v17 mRNA表达差异无统计学意义(P>0.05);乳腺癌临床Ⅲ期CD44v17 mRNA表达(4.18±0.92)明显高于Ⅰ期(2.73±0.97),Ⅱ期(2.76±0.96)(P<0.05);乳腺癌CD44v17 mRNA表达与雌激素受体、孕激素受体(progesterone receptor,PR)和cerbB-2表达无明显相关性(P>0.05)。结论:CD44v17 mRNA在乳腺癌组织中高表达,在乳腺癌的发生发展中起重要作用,可能与乳腺癌的侵袭、转移及预后有关。Objective: To investigate the expression and clinical significance of the new CD44 variant(CD44v17) in breast cancer tissues.Methods: The specific primers of the new CD44 variant(CD44v17)(GeneBank NO.FJ216964) found in MCF-7/Adr in our laboratory were designed.The expression of CD44v17 at mRNA levels in 52 cases of breast cancer tissues and corresponding normal tissues was detected with the SYBR GreenⅠreal time fluorescent quantitative PCR(qRT-PCR) method according to standard curve of CD44v17 mRNA.The relationship between the CD44v17 mRNA expression and biological characteristic of breast cancer was evaluated.Results: The significant difference of the CD44v17 mRNA expression levels was found between the group of breast cancer tissues(3.10±1.12) and the control group(2.31±0.84)(P0.01).In addition,The expression levels of CD44v17 mRNA in the group of lymph nodes metastasis(4.07±1.13) were higher than that without lymph nodes metastasis(2.75±0.89)(P0.01);CD44v17 mRNA expression in the 5 cm group(4.18±0.95) was significantly higher than that in the ≤5 cm group(2.96±1.07)(P0.05);CD44v17 mRNA expression in the carcinoma medulla group(5.27±0.30) was higher than in the intraductal carcinoma(3.28±1.08)and infitrating ductal carcinoma group(2.92±1.04)(P0.05);but there was no significant difference in CD44v17 mRNA expression between the intraductal carcinoma and infitrating ductal carcinoma group(P0.05).CD44v17 mRNA expression in Ⅲ clinical stage(4.18±0.92) was significantly higher than that in Ⅰ(2.73±0.97)or Ⅱ group(2.76±0.96)(P0.05);however,CD44v17 mRNA expression and estrogen receptor(ER),progesterone receptor(PR) or cerbB-2 did not exist statistical significance(P0.05).Conclusion: CD44v17 mRNA was over expressed in breast cancer.The expression of CD44v17 mRNA may play important roles in tumorigenesis and progression of breast cancer.The expression of CD44v17 mRNA may be correlated with invas
