HPLC法同时测定珠黄吹喉散中儿茶素和盐酸小檗碱的含量

摘　　要：目的建立同时测定珠黄吹喉散中儿茶素和盐酸小檗碱含量的方法。方法采用HPLC法,色谱柱:AgilentHypersilC18柱(250mm×4.6mm,5μm),流动相:乙腈-0.1%磷酸溶液梯度洗脱,检测波长:270nm,流速:1.0mL/min,进样量:10μL。结果儿茶素在0.0240～0.4800μg范围内与峰面积呈现良好的线性关系,r=0.9998;平均回收率为98.65%,RSD=1.10%(n=6);盐酸小檗碱在0.0132～0.2640μg范围内与峰面积呈现良好的线性关系,r=0.9997;平均回收率为97.87%,RSD=1.17%(n=6)。结论该方法准确、重复性好,可用于珠黄吹喉散的质量控制。Objective To establish a method to determine the contents of catechin and berberine hydrochloride in Zhuhuang Chuihou Powder simultaneously.Methods HPLC was applied with the chromatographic condition as follows：Agilent Hypersil C18（4.6 mm×250 mm,5μm）,ACN-0.1%H3PO4 as the mobile phrase,gradient elution,detection wavelength being 270 nm,flow rate being 1.0 mL·min-1 and injection volume being 10μL.Results The calibration curves were linear within the range of 0.024 0～0.480 0μg for catechin and 0.013 2～0.264 0μg for berberine hydrochloride,with the average recovery being 98.65%（RSD=1.10%）,r=0.999 8 for catechin and 97.87 %（RSD=1.17%）,r=0.999 7 for berberine hydrochloride.Conclusion This accurate and reproducible method can be used to control the quality of Zhuhuang Chuihou Powder.
