摘  要:目的:探讨造血细胞磷酸酶(SHP-1)基因和天冬酰氨特异酶切的半胱氨酸蛋白酶(caspase-1、3)基因表达与急性白血病(acute leukemia,AL)患者的化疗效果的关系。方法:用RT-PCR方法检测急性白血病患者骨髓单个核细胞中SHP-1和caspase-1、3的mRNA表达。结果:AL患者SHP-1和caspase-1、3表达率分别为33.3%、80.0%、100.0%;SHP-1和caspase-1、3基因阳性患者的首次完全缓解(CR)率分别为85.0%、64.3%和55.8%,明显高于阴性患者(分别为15.6%、20.0%),差异均有统计学意义(P<0.05);caspase-1、3与SHP-1表达呈正相关。结论:SHP-1可能是一个潜在的抑癌基因,它可能是通过激活下游的凋亡基因caspase-1、3发挥作用;caspase-1、3与SHP-1基因表达呈正相关且与患者的CR率有关,表明它们均参与了急性白血病的转归,可同时作为判断急性白血病患者的疗效和预后指标。Objevtive To explore the association of the expressions of hematopoietic cell phosphatase (SHP-1) gene and cysteinyl aspartate-specific proteinases (caspase-1,and -3) genes with the efficacy of chemotherapy in patients with acute leukemia. Methods The expressions of SHP-1 and caspase-1 and -3 genes in bone marrow-derived mononuclear cells were detected in the patients by reverse transcriptase-polymerase chain reaction (RT-PCR). Results The expression rate of SHP-1 and caspase-1,and -3 genes was 33.3%,80.0%,and 100.0%,respectively. The initial complete remission (CR) was 85.0% in SHP-1 positive patients,64.3% in caspase-1 positive patients,and 55.8% in caspase-3 positive patients,and was obviously higher than those in the patients with negative SHP-1,caspase-1 or -3 (P 0.05). SHP-1 was positively related with caspase-1 and -3. Conclusions SHP-1 gene may be a potential anti-oncogene and may play its role by activating caspase-1 and -3 genes. The expressions of caspase-1 and -3 are positively related with SHP-1 expression. All of the genes are associated with complete remission,suggesting that they participate in the prognosis of leukemia. They could be used as indicators for evaluating the therapeutic efficacy and judging prognosis in acute leukemic patients.
