升气血口服液对K562细胞增殖的影响

摘　　要：目的:研究升气血口服液对人慢性髓系白血病细胞株K562细胞增殖的影响。方法:应用MTT法检测升气血口服液对K562细胞增殖的作用。Hoechst33258荧光染色、透射电镜观察细胞形态学变化。结果:MTT结果表明升气血口服液能抑制K562细胞增殖,且呈时间依赖性。Hoechst33258荧光染色发现染色质凝集,着色深而呈亮蓝色,部分细胞核呈分叶、碎片状或边集。透射电镜观察升气血口服液作用的K562细胞胞质出现大量空泡变性,细胞核凝集等细胞凋亡形态学改变。结论:升气血口服液对K562细胞具有抑制增殖和诱导凋亡作用。Objective：To study the proliferation effects of oral liquid of lifting QI-blood on human leukemic K562 cells.Methods：Cell proliferation effect of oral liquid of lifting QI-blood on K562 cells was evaluated by MTT assay.The morphological changes were observed by Hoechst33258 fluorescein stain and electron microscope.Results：MTT assay showed that oral liquid of lifting QI-blood inhibit proliferation of K562 cells in a time-dependant manner.Hoechst33258 fluorescein stain appear chromatic agglutination and have bright blue nuclear.Part of nuclear submit sublobe,chip or edge collection.Morphological changes of apoptosis about cytoplasm vacuolar degeneration and nuclear condensation were observed by electron microscope in oral liquid of lifting QI-blood-treated cells.Conclusion：Oral liquid of lifting QI-blood inhibit proliferation and induce apoptosis on K562 cells.
