全反式维甲酸对兔后发性白内障的抑制作用

摘　　要：目的探讨全反式维甲酸(ATRA)对兔后发性白内障的抑制作用。方法将12只健康家兔(24只眼)随机分为眼用平衡盐液组(空白对照组)和ATRA组(药物实验组),每组6只兔(12只眼),在全麻下进行透明晶状体囊外摘除术。分别于术后1、3、7、14、30和60 d对术眼进行裂隙灯和眼底检查,并抽取房水置于-80℃保存。用酶联免疫吸附测定术后兔眼房水中的白细胞介素-1β(IL-1β)和转化生长因子-β(TGF-β)的含量。3月后处死动物,分别对两组兔眼进行组织病理学及电镜观察。结果术后房水中IL-1β和TGF-β含量实验组明显低于对照组(P<0.001)。术后3月实验组晶体后囊透明度优于对照组(P<0.05)。对照组晶体后囊前覆盖一层晶体上皮细胞,实验组晶体后囊前无明显晶体上皮细胞增殖。结论 ATRA可抑制房水中IL-1β和TGF-β的含量,能够有效抑制后发性白内障的发生。Objective To study the inhibitory effects of all-trans retinoic acid （ATRA） on posterior capsule opacification of rabbits. Methods Extracapsular cataract extraction was performed on 12 rabbit eyes which were divided into two groups： control, ATRA group. The control group was injected balanced salt solution in the operation, while the ATRA group was injected ATRA. The aqueous humors were collected at 1, 3, 7, 14, 30 and 60 days after operation and determined the content ration of interleukin-1β （IL-1β） and transform growth factor-β （TGF-β） by enzymelinked immunosorbent assay. Clinical observation was carried out regularly. Three months later, the biomicroscopic photograph of posterior capsule and the pathological changes of posterior lens capsule were observed by microscopy. Results The content of IL-1β and TGF-β in ATRA group was significantly lower than control group, showing a statistically significant difference （P 〈0.001）. In 3 month after the operation, the eye numbers developed posterior capsule opacification （PCO） were 12 and 4 in control group and ATRA group, with a significant difference between control group and ATRA group （P 〈0.05）. No or only sporadic lens epithelial cells and fibrin sticking were found at posterior capsule of lens in ATRA group under the light and electron microscope, and severe PCO was seen in control group. Conclusion The implantation of ATRA into the anterior chamber may to some extent inhibit the occurrence of after-cataract.
