摘  要:目的观察大鼠局灶性脑梗死模型中尿激酶(UK)延迟溶栓对脑梗死体积和小胶质细胞增殖的影响,探讨小胶质细胞对脑梗死的作用及可能机制。方法制作大鼠大脑中动脉梗死(MCAO)模型,随机分成4组:Sham组、NS组、UK1.5组和UK6组。术后24h分别用TTC染色、HE及免疫组化检测脑梗死体积、病理变化及小胶质细胞增殖。结果 Sham组无梗死;UK1.5组与NS及UK6组相比,梗死缩小(P<0.01);UK6组与NS组无差异(P>0.05)。免疫组化,植物凝集素在各缺血组的表达均增加(P<0.01);与NS及UK6组相比,UK1.5组表达减少(P<0.01);UK6组与NS组无差异(P>0.05)。结论尿激酶延迟溶栓不能抑制脑梗死后小胶质细胞的增殖活化,也无法缩小梗死体积,反而增加发生并发症的风险,有可能加重缺血区脑组织损害。Objective To approach the effects of microglia and its possible mechanism after delayed urokinase thrombolysis on experimental focal cerebral infarction in rats.Methods Rats were subjected to middle cerebral artery occlusion.Rats were randomly divided into four groups:Sham group,NS group,UK1.5 group and UK6 group.The infarct volume of brain was investigated by TTC coloring.The proliferation of microglia was analyzed by immunohistochemistry 24 h after operation.Results Cerebral ischemia could not be seen in Sham group.Infarct volume decreased in UK1.5 group compared with NS and UK6 group(P〈0.01);UK6 group,compared with NS group,was no significant difference(P〉0.05).Immunohistochemistry revealed that the expression of phytagglutinin was increased in all ischemic groups(P〈0.01).The expression was weaker in UK1.5 group than NS and UK6 group(P〈0.01);and it was no significant difference in UK6 and NS group(P〉0.05).Conclusion Delayed thrombolysis with UK not only failed to inhibit the proliferation of microglia after focal cerebral infarction so that infarct volume can not be reduced,but increased the occurrence of complications and was likely to aggravate brain tissue damage in the ischemic region.
