摘 要:目的探讨肿瘤转移抑制基因K ISS-1及基质金属蛋白酶9(MMP-9)与胃癌侵袭、转移的关系,为研究胃癌的转移机制及治疗提供理论基础。方法采用逆转录聚合酶链反应(RT-PCR)检测36例胃癌组织及36例正常胃组织中K ISS-1 mRNA及MMP-9 mRNA的表达情况,分析其与胃癌患者各临床病理指标的关系及二者的相关性。结果 K ISS-1 mRNA在胃癌组织中的阳性表达率及表达水平均低于正常胃组织(P均<0.01),并且其低表达与淋巴结转移密切相关(P<0.05);MMP-9 mRNA在胃癌组织中的阳性表达率及表达水平均高于正常胃组织(P均<0.05),MMP-9 mRNA的高表达与癌的浸润深度和淋巴结转移密切相关(P均<0.05);K ISS-1与MMP-9表达呈负相关(P<0.05)。结论 K ISS-1表达缺失和MMP-9过表达可能与胃癌的侵袭相关。Objective To explore relationship between metastasis suppressor gene KISS-1,matrix metelloproteinases 9(MMP-9) and the invasion and metastasis of gastric carcinoma,then to provide a theoretical foundation for investigation of metastatic mechanism of gastric carcinoma.Methods Reverse transcription-polymerase chain reaction(RT-PCR) method was used to detect KISS-1 mRNA and MMP-9 mRNA expression in 36 cases of gastric carcinoma and 36 cases of normal gastric epithelia.Results The positive expression rate and relative level of KISS-1 mRNA in gastric carcinoma were significantly lower than those in normal gastric mucosa(P0.01).The low expression of KISS-1 mRNA in gastric carcinoma correlated with lymph node metastasis(P0.05).The positive expression rate and relative level of MMP-9 mRNA in gastric carcinoma were significantly higher than those in normal gastric mucosa(P0.05).The high expression of MMP-9 mRNA in gastric carcinoma correlated with the depth of invasion and the metastasis of lymph node(P0.05).The expression of KISS-1 mRNA had negative correlation with the expression of MMP-9 mRNA.Conclusion Low-expression of KISS-1 and over-expression of MMP-9 may be corrrelated with invasion of gastric carcinoma.
