摘 要:目的合成新化合物过氧化甲酰胺,对其合成工艺进行优化,并观察小鼠对它的急性毒性反应。方法采用正交试验法,以合成过氧化甲酰胺的产率为指标进行试验。通过排水集气法收集氧气。以过氧化甲酰胺的最大浓度给小鼠灌胃给药,进行急性毒性试验。结果最佳合成工艺为:用过氧化氢和甲酰胺质量比5∶2,在(10±2)℃水浴温度下反应后,冷藏时间24 h。过氧化甲酰胺实际产氧量为148.0 ml/g。急性毒性试验未出现小鼠死亡,未测出LD50,过氧化甲酰胺的最大给药量为5.64 g/kg。结论该合成工艺合理,产率高。合成的过氧化甲酰胺产氧量高,具有较好的安全性。Objective To synthetize a new compound, formamide peroxide, and to optimize its synthesis technology, then to observe the acute toxicity reaction in mice. Methods The synthesis process of for- mamide peroxide was optimized using orthogonal test, and the production rate of synthetical formamide peroxide was used as the index. The oxygen was collected by gas-collecting method of drainage. In acute toxicity test, the mice were intragastrically administered with maximum concentration of formamide perox- ide. Results The best synthesis process was as follows: the mass ratio of hydrogen peroxide to formamide was 5:2, and the reaction conditions were in (10 + 2)℃ water bath following 24 hours of cold storage. The actual oxygen production of formamide peroxide was 148.0 ml/g. In acute toxicity test, none of dead mice was found, so the median lethal dose could not be obtained; the largest dosage of formamide peroxidation was.5.64 g/kg. Conclusion The synthesis technology is reasonable with high yield. The synthetical formamide peroxide has high oxygen production with good security.
