摘 要:目的研究筛选蜗牛酶降解壳聚糖的最佳反应条件。方法采用正交试验法,以降解产物中寡糖的量和糖收率为指标,考察温度、酶浓度、pH对蜗牛酶降解壳聚糖的影响,筛选最佳反应条件;采用SDS-PAGE检测降解产物,用改良Schales法测定还原糖的含量并计算寡糖的收率。结果蜗牛酶降解壳聚糖的最佳反应条件为:pH4、40℃、酶底比为8:100。最佳条件下的寡糖收率为64.74%,产物主要是聚合度为4以上的寡糖。结论在最佳反应条件下,蜗牛酶能较好地降解壳聚糖。OBJECTIVE To investigate the optimal reaction conditions for chitosan hydrolysis with snailase.METHODS The optimal conditions were screened with orthogonal test,according to the chitooligosaccharide content in the production and the chitooligosaccharide yield,and the effect of temperature,enzyme concentration and pH on chitosan hydrolysis with aid of snailase were investigated.SDS-polyacrylamide gel electrophoresis was used to analyzed the hydrolysis products and a modified-Schales method was used to determine the chitooligosaccharide content.The chitooligosaccharide yield was calculated.RESULTS The optimal conditions for chitosan hydrolysis with snailase were as follows:pH4.0,the temperature of 40 ℃,the ratio of the enzyme to substrate:8:100.The chitooligosaccharide yield was 64.74% and the polymerization degree of the major hydrolysis products was above 4.CONCLUSION Chitosan can be well hydrolysed by snailase under the optimal conditions.
