摘　　要：[目的]研究手性药物氨甲喋呤(MTX)单一对映体对人肝癌细胞株HepG-2的体外增殖的抑制作用。[方法]应用MTT比色法检测MTX对映体[(+)MTX、(-)MTX]对HepG-2细胞增殖抑制作用,用倒置显微镜观察细胞的形态学变化。[结果]0.1～150μmol/L(+)MTX和10～150μmol/L(-)MTX可抑制HepG-2细胞的生长,且其作用具有剂量和时间依赖性(P<0.01),但抑制强度为(+)MTX>(-)MTX;倒置显微镜观察不同浓度(+)MTX和(-)MTX作用HepG-2细胞株不同时间后,细胞出现不同程度的形态学改变。[结论](+)MTX和(-)MTX对HepG-2细胞的抑制作用不同,(+)MTX抗增殖作用明显强于(-)MTX。[ Purpose] To study the inhibition of methotrexate（MTX） enantiomers on proliferation of human Hepatoma HepG-2 cell line. [Methods] The inhibition of MTX enantiomers [（＋）MTX, （-）MTX] on proliferation of HepG-2 cells were detected by the MTT assay. The morphological profile were examined by inverted microscopy. [Results] Both （＋）MTX dosed 0.1-150μmol/L and （-）MTX dosed 10-150μmol/L inhibited the growth of HepG-2 cells in a time- and dose-dependent manner （P〈0.01）.The inhibition of （＋）MTX was stronger than that of （-） MTX. When the different concentration of MTX enantiomers were administrated at the different times, the different morphologies of HepG-2 cells were found. [ Conclusions] The inhibition of （＋）MTX and （-）MTX on HepG-2 cells was different and the anti-proliferation of （＋）MTX was significantly stronger than that of （-）MTX.
