顺铂联合葛根素纯品和葛根粗提物对H446细胞增殖及Bax、Bcl-2蛋白表达的影响

摘　　要：目的:观察顺铂(DDP)单独及联合葛根素纯品(SP)和(或)葛根粗提物(CP)对小细胞肺癌H446细胞增殖及Bax、Bcl-2蛋白表达的影响。方法:取对数生长期的H446细胞,随机分为16组,运用噻唑蓝法测定不同质量浓度DDP(1.0、2.0、4.0和8.0mg/L)单独及联合80mg/LSP和(或)CP对H446细胞的增殖抑制率,运用免疫细胞化学染色法测定16组细胞中Bcl-2和Bax蛋白的表达,运用析因设计方差分析法分析检测结果。结果:析因设计方差分析结果显示:不同质量浓度DDP(F=3973.386,P<0.001)、SP(F=468.017,P<0.001)和CP(F=789.757,P<0.001)对H446细胞的增殖抑制不同,随着DDP质量浓度的增高,联合80mg/LSP和80mg/LCP对H446细胞的抑制越明显;DDP联合SP(F=70.686,P<0.001)、DDP联合CP(F=87.191,P<0.001)及DDP联合SP和CP(F=28.471,P<0.001)对H446细胞增殖抑制作用增强。DDP、SP和CP上调Bax蛋白的表达,下调Bcl-2蛋白的表达。结论:联合应用SP、CP能增强DDP对H446细胞的增殖抑制作用,该作用可能与上调Bax蛋白表达、下调Bcl-2蛋白表达有关。Aim：To compare the effects of cisplatin （DDP） alone or combined with pure puerarin （SP） and pueraria crude extract （CP） on the growth and expression of Bax and Bcl-2 of H446 cells.Methods：H446 cells in log phase were selected and divided into 16 groups randomly,MTT assay was used to determine the depressant effect of different concentrations of DDP（1.0,2.0,4.0 and 8.0 mg/L） alone or combined with SP and CP（80 mg/L） on the proliferation of H446 cells. Immunohistochemistry were used to determine the expression of Bcl-2 and Bax protein in 16 groups. The results were analyzed by factorial design variance analysis.Results：The results of factorial design variance analysis showed that different concentrations of DPP（F=3 973.386,P0.001）,SP（F=468.017,P0.001）,and CP（F=789.757,P0.001） had different inhibition ratios to H446 cells,and the higher the concentrations of DPP,the more obvious inhibition of 80 mg/L SP and CP to H446 cells. DDP combined with SP（F=70.686,P0.001）,CP（F=87.191,P0.001）,SP and CP（F=28.471,P0.001）could strengthen DDP＇s inhibition effect on the growth of H446 cell. DDP,SP and CP could up-regulate the expression of Bax protein,and down-regulate the expression of Bcl-2 protein.Conclusion：DDP combined with CP and SP respectively could enhance DDP＇s inhibition on H446 cells,this effect may attribute to the up-regulation of Bax and down-regulation of Bcl-2 protein.
