摘  要:目的:观察骨形成蛋白7重组腺病毒(Ad-BMP7)转染兔软骨细胞后BMP7的表达。方法:分离培养3周龄兔膝关节软骨细胞,将Ad-BMP7转入,RT-PCR检测BMP7在mRNA水平的表达,Western Blot和免疫组化染色检测其在蛋白水平的表达。另设空白对照组。结果:实验组在mRNA水平和蛋白水平均能有效表达BMP7基因。结论:携带BMP7基因的重组腺病毒体外转染的关节软骨细胞,可以产生BMP7。Objective:To investigate the expression of BMP7 in cultured rabbit chondrocytes with BMP7 gene transfected by adenovirus vector(Ad-BMP7) in vitro.Methods:Chondrocytes were isolated and cultured from a 3-week-old rabbit knee. Ad-BMP7 was transfected into the cells at the doses of 50 and 100 MOI.BMP7 mRNA expression was observed by RT-PCR and protein expression was evaluated by Western blot and immunohistochemistry.Control groups were transfected with empty adenovirus vector.Results:BMP7 mRNA expressed in experimental groups could be observed by RT-PCR. BMP7 protein could be detected in the chondrocytes by Western blot.The strong positive signal of immunohistochemistry staining for BMP7 transfected chondrocytes could be detected in experimental groups,while in the control groups weak positive immunohistochemistry staining was exhibited.Conclusion:The results suggest that chondrocytes transfected with Ad-BMP7 could produce BMP7.
