侧脑室微量注射瘦素对OEP大鼠子宫腔液蛋白表达的影响

摘　　要：目的:探讨瘦素通过下丘脑-垂体轴对子宫的调节作用。方法:应用双向凝胶电泳(2-DE)技术分离OEP大鼠侧脑室微量注射瘦素及生理盐水6h后的子宫腔液总蛋白,图像分析识别差异表达的蛋白质点,基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)对差异表达的蛋白质点进行鉴定。结果:与生理盐水对照组相比,瘦素实验组有6个差异表达蛋白质,其中5个蛋白点升高,1个蛋白点降低。经过质谱分析结合数据库检索,获得有意义的蛋白点,上调蛋白点为补体B因子、冠蛋白、肌动蛋白结合蛋白、丝氨酸蛋白酶抑制剂B1、烯醇酶,下调蛋白点为补体C3。结论:这些差异表达蛋白涉及炎症反应、细胞分化、细胞迁移、细胞凋亡等功能,瘦素可能通过下丘脑-垂体轴从免疫、生殖等方面对子宫发挥调节作用。Objective：To study a potential role for leptin in the regulation of uterus through hypothalamo-pituitary axis.Methods：The total proteins of the uterine luminal fluid（ULF）of ovariectomized estrogen-primed（OEP）rats injected with physiologic saline and leptin through the lateral cerebral ventricle for 6 hours were separated by two-dimensional gel electrophoresis（2-DE）.Imaging analysis was conducted to distinguish the differentially expressed protein spots of ULF,and the proteins were identified by matrix-assisted laser desorption/ionization time of flight mass spectrometry（MALDI-TOF-MS）.Results：Six differential expression proteins were confirmed,five with high expression and one with low expression.By sequence database searching,the proteins with high expression were complement B factor,coronin,serine（or cysteine）proteinase inhibitor,clade B,enolase 1,and the protein with low expression was complement component 3.Conclusion：These proteins are involved in inflammatory reaction,cell differentiation,cell migration,apoptosis,and so on.Leptin plays an important role in regulating immunoregulatory and reproductive function of the uterus through the route of hypothalamo-pituitary axis.
