摘  要:目的探讨应用细胞CD55高表达的特性替代Hoechst33342染色来分离乳腺癌SP细胞的可行性。方法应用Hoechst 33342及抗CD55抗体对两种乳腺癌细胞株进行荧光染色,应用FACS-Vantage和FACSVantageSE分选SP和MP细胞、CD55高表达(CD55hi)和CD55低表达(CD55lo)细胞,对比2组细胞对血清缺失诱导的凋亡的耐受性及CD55hi和CD55lo细胞的克隆形成能力。结果对分选的CD55hi和CD55lo细胞行Hoechst 33342染色,2组细胞分别位于SP和MP区域中。在血清缺失情况下,SP细胞和CD55hi细胞分别比MP细胞和CD55lo细胞对凋亡刺激具有更高的耐受性。CD55hi细胞较CD55lo细胞具有更强的克隆形成能力。结论细胞CD55高表达的特性可以替代Hoechst33342染色用于分离乳腺癌SP细胞。Objective To investigate whether CD55 overexpression may be employed to isolate side-population (SP)cells from mammary carcinoma cells instead of Hoechst 33342 staining. Methods The two kinds of mammary carcinoma cell lines were stained with Hoechst 33342 or anti-CD55 antibody respectively,then sorted as SP or main-population(MP)cells,CD55hi or CD55lo cells with FACS-Vantage and FACS Vantage SE. The tolerance to apoptosis induced by serum depletion was compared between these two groups,and the colony formation between CD55hi and CD55lo cells. Results When cells were stained with Hoechst 33342 again,the sorted CD55hi and CD55lo cells were distributed in SP and MP regions respectively. The proportion of viable cells was higher in SP or CD55hi cells than that in the counterparts. Colony number of CD55hi cells was significantly higher than that of CD55lo cells. Conclusion The CD55 overexpression can be employed to isolate side-population cells from mammary carcinoma cells instead of Hoechst 33342 staining.
