摘  要:目的:了解肝纤维化发展和逆转过程中RECK基因表达与基质金属蛋白酶-2(MMP-2)活性之间的相关性。方法:SD雄性成年大鼠随机分为对照组和肝纤维化组,用CCl4皮下注射建立肝纤维化模型。8周后,肝纤维化组随机分为肝纤维化进展组和肝纤维化自然逆转组,前者继续予CCl4注射,后者停用CCl4。采用天狼猩红染色判断肝纤维化程度,明胶酶谱法检测肝组织MMP-2的活性,RT-PCR法检测RECK基因的表达情况。结果:MMP-2的活性随肝纤维化程度的加重而逐渐升高,而肝纤维化逆转时则随时间推移MMP-2的活性逐渐下降;而RECK的变化相反,RECK表达和MMP-2的活性呈负相关(P<0.05)。结论:在大鼠肝纤维化发展和逆转过程中,RECK表达与MMP-2活性可能有密切关系,前者对后者可能起抑制作用。Objective:To investigate the relation between RECK expression and matrix metalloproteinase-2(MMP-2) activity in the development and the reversion process of hepatic fibrosis in rats.Methods:Spraque Dawley male rats were randomly divided into control group and hepatic fibrosis group.The hepatic fibrosis model was induced by CCl4 in the experiment group.Eighth weeks later, the hepatic fibrosis group were randomly divided into two groups,including hepatic fibrosis development group which were keep on induced by CCl4 and hepatic fibrosis reversion group which were stopped using CCl4.The collagen deposition,MMP-2 activity and RECK expression in the liver tissues were detected by Sirius red staining,zymography and RT-PCR respectively.Results:It was found that MMP-2 activity increased gradually in the development process of the hepatic fibrosis,and descended in the reversion process.But RECK expression was reversed change at the same time.And negative correlation between RECK expression and MMP-2 activity in the liver tissues(P〈0.05).Conclusions:MMP-2 activity and RECK expression may be closely related in the development and reversion process of hepatic fibrosis.RECK maybe have an depressant effect on MMP-2 activity.
