摘 要:目的:观察人参皂苷Rb1与红景天苷对中波紫外线(ultraviolet B,UVB)辐照后人角质形成细胞光产物水平的影响及人参皂苷Rb1和红景天苷光保护作用机制。方法:使用含10%小牛血清的RMPI-1640培养液培养人角质形成细胞HaCaT,定量(60 J.cm-2)UVB照射细胞,MTT法检测细胞增殖活性;培养液中加入人参皂苷Rb1和红景天苷,测定UVB照射细胞后细胞中超氧化物岐化酶(SOD)、谷光甘肽(GSH)、氧化氢酶(CAT)活性以及丙二醛(MDA)含量的变化。结果:在暴露于UVB后,经人参皂苷Rb1和红景天苷处理,细胞内抗氧化酶SOD、GSH、CAT的活性提高,MDA的产生减少,与未用药组比较差异有统计学意义(均P<0.01)。结论:人参皂苷Rb1和红景天苷具有增加人角质形成细胞内抗氧化酶活性、抑制脂质过氧化反应及抗氧化作用,可以用于对抗皮肤光老化。Objective:To observe the effect of ginsenoside Rb1 and salidroside on the production of human keratinocytes after exposed under UVB,and explore its mechanism of protection for the photodamage.Methods:Human keratinocytes HaCaT were cultured in RMPI-1640 with 10% fetal bovine serum and exposed to certain dosage(60 J·cm^-2) of UVB,the growth activity was evaluated by MTT,then ginsenoside Rb1 and salidroside were added into culture medium separately,the activity of SOD,GSH,CAT and content of MDA in cells were evaluated.Results:The activity of SOD,GSH,CAT were increased and the content of MDA was decreased after treated by ginsenoside Rb1 and salidroside.There was statistical significant difference between the medicine treated groups and untreated control groups(all P〈0.01).Conclusion:Ginsenoside Rb1 and salidroside posses the ability of anti-oxide by means of increasing the intro-cellular activity of antioxidase,and inhibiting the reaction of lipid peroxidation.These two medicine agents can be used in anti-skin photoaging.
