MG132对体外培养人晶状体上皮细胞增殖的抑制作用

摘　　要：目的研究蛋白酶体抑制剂MG132对晶状体上皮细胞系SRA01/04细胞增殖的影响,探讨其抑制细胞增殖的有效药物浓度及分子机制。方法不同浓度的MG132分别处理SRA01/04细胞36h,通过MTT法检测MG132对SRA01/04细胞增殖的影响,通过流式细胞术检测MG132对SRA01/04细胞凋亡的影响,通过Annexin V-FITC/PI双染法在荧光显微镜下观察SRA01/04细胞早期凋亡形态。结果随着MG132浓度的升高,对SRA01/04细胞增殖的抑制作用逐渐增强,半数有效抑制浓度IC50为2.48μmol·L-1。MG132可诱导SRA01/04细胞凋亡;流式细胞术结果表明:浓度为2.5μmol·L-1、5.0μmol·L-1的MG132作用SRA01/04细胞36h后,细胞早期凋亡率分别为6.55%±0.35%和13.75%±3.18%,而0μmol·L-1为0.75%±0.21%,差异均有统计学意义(均为P<0.05)。凋亡细胞被Annexin V-FITC单独染色(绿色荧光),活细胞不被染色,坏死细胞被Annexin V-FITC和PI(红色荧光)同时染色。结论蛋白酶体抑制剂MG132可通过诱导细胞凋亡来抑制SRA01/04细胞的增殖,起到防治后发性白内障的作用。Objective To investigate the effective drug concentration and mechanisms of proteasome inhibitor MG132 on inhibiting proliferation of human lens epithelial cell line SRA01/04.Methods SRA01/04 cells were treated with MG132 at different concentrations for 36 hours.The effects of MG132 on the proliferation of SRA01/04 cells were analyzed with MTT assay,effects of MG132 on the apoptosis of SRA01/04 cells were analyzed by flow cytometry,and early apoptotic morphologies of SRA01/04 cells were observed after Annexin V-FITC/PI double staining by fluorescence microscope.Results The inhibitory effect of MG132 on SRA01/04 cells proliferation enhanced with the increase of the concentration of MG132.The 50% inhibitory concentration of MG132 was 2.48 μmol·L-1.MG132 could induce the apoptosis of SRA01/04 cells.The early apoptotic rates of SRA01/04 cells were 6.55%±0.35% and 13.75%±3.18% after being treated with MG132 at concentrations of 2.5 μmol·L-1 and 5.0 μmol·L-1 for 36 hours by flow cytometry,respectively,but 0.75%±0.21% at concentration of 0 μmol·L-1.There were statistical differences（all P0.05）.Apoptotic cells were stained only by Annexin V-FITC（green fluorescence）,living cells were not stained.Necrotic cells were stained by Annexin V-FITC and PI（red fluorescence） double staining.Conclusion Proteasome inhibitor MG132 can inhibit the proliferation of SRA01/04 cells by inducing cell apoptosis,and plays a role in the prevention of posterior capsular opacification.
