摘 要:目的:探讨大黄素是否可通过影响淋巴细胞内活性氧的产生,促进淋巴细胞的凋亡来发挥其免疫抑制作用。方法:分别提取健康成年人外周静脉血中淋巴细胞,建立混合淋巴细胞培养(MLC)模型,给予不同浓度的大黄素,并进行不同时间点干预,利用流式细胞技术检测淋巴细胞内活性氧的产生水平及淋巴细胞凋亡情况,并给予还原性谷胱甘肽清除细胞内活性氧,流式细胞技术再次观察细胞内活性氧水平及淋巴细胞凋亡情况。结果:在高、中、低浓度组,大黄素与混合培养淋巴细胞共培养24h、48h、72h后,淋巴细胞内活性氧产生水平增高,在1μmol/ml-100μmol/ml的浓度范围内,不同浓度大黄素组淋巴细胞凋亡率与对照组相比均有不同程度的升高(P<0.05),并呈现浓度和时间依赖性。结论:大黄素可能通过提高淋巴细胞内活性氧产生水平,进而促进淋巴细胞凋亡来发挥其免疫抑制作用。Objective:To investigate whether emodin can affect the expression of reactive oxygen species in lymphocyte, promoting lymphocyte apoptosis to play immunosuppressive effects. Methods: The modle of human mixed lymphocyte culture was applied and lymphocytes were isolated from different healthy adult volunteers with peripheral blood samples. Emodin was administered with different dose in different culture time. The reactive oxygen spieces in lymphocytes and the lymphocyte apoptosis with or without administration of glutathione were detected by flow cytometry. Results: Within the concentration from 1 umoL/ml to 100umol/ml, the concentration of reactive oxygen spieces in lymphocytes and the radio of lymphocyte apoptosis were increased with the administration of Emodin in a dose - dependent fashion ; the ratio of lymphocyte apoptosis decreased with the administration of glutathione. Conclusion: Emodin may attain the effect of immunosuppression through inhibiting lymphocyte proliferation by elevating the concentration of reactive oxygen spieces in lymphocytes and the level of lymphocyte apoptosis.
