摘 要:目的 探讨FKHR、AKT蛋白在大鼠蛛网膜下腔出血脑皮质中的表达及意义.方法 将24只大鼠随机分为3组:假手术组、蛛网膜下腔出血(SAH)组、SAH+尼莫地平组,每组8只.采用枕大池二次注血法建立Wistar大鼠蛛网膜下腔出血模型,Loeffler法进行大鼠神经行为学评分,Western blotting检测脑皮质中FKHR(FOXO1)、P-FKHR、AKT及P-AKT蛋白的表达.结果 神经行为评分与假手术组(4.81±0.06)相比,SAH组(2.49±0.34)的神经行为评分明显降低(P〈0.05),FKHR的表达明显增加(P〈0.01),AKT的表达变化不明显,P-AKT/AKT及P-FKHR的表达明显下降(均P〈0.01),而给予尼莫地平治疗后,神经行为评分明显升高(P〈0.01),P-AKT/AKT及P-FKHR的表达亦升高(均P〈0.01).结论 P-AKT及P-FKHR参与了大鼠蛛网膜下腔出血所致的损伤过程,而尼莫地平通过上调P-AKT及P-FKHR的减轻SAH所致脑损伤.Objective To study the significance and expression of FKHR and AKT after subarachnoid hemorrhage (SAH) in rat brain cortex. Methods Twenty-four rats were randomly divided into three groups; sham, SAH and SAH plus nimodipine (n =8 each). A reliable SAH model was established by double injections of blood into cistern magna in Wistar rats. The neurological scores were measured by Loeffler and the expressions of FKHR, P-FKHR, AKT and P-FKHR detected by Western blot. Results Compared with sham group, the neurological score of SAH group obviously decreased (P 〈 0.05), the expression of FKHR became elevated in rat cortex (P 〈 0.01), the expression of AKT had no change and the expressions of P-AKT and P-FKHR obviously decreased (all P 〈0. 01). But the neurological score markedly increased (P 〈0. 01) and the expressions of P-AKT and P-FKHR became elevated (all P 〈0. 01) after administration of nimodipine. Conclusion Both P-AKT and P-FKHR are involved in the process of brain cortex damage induced by SAH. The protective effects of nimodipine on brain injury induced by SAH may be related to the elevated expressions of P-AKT and P-FKHR in brain cortex.
