摘  要:目的探讨肝癌组织miR-122的表达及其与肝癌细胞周期调控的关系。方法采用原位杂交法检测26例肝癌组织miR-122的表达。用流式细胞仪检测肝癌细胞周期。结果肝癌组织miR-122阳性表达率为26.92%,而癌旁组织为80.77%,两组之间差异有统计学意义(P<0.01)。与癌旁组织相比,肝癌细胞S期比例升高,G1-G0期比例降低(P<0.05);与miR-122阳性组比较,miR-122阴性组肝癌细胞S期比例升高,G1-G0期细胞比例亦降低(P<0.01)。结论肝癌细胞miR-122的表达下调,促进癌细胞的分裂,使癌细胞具有更强的增殖、侵袭和转移的能力。Objective To investigate the expression of miR-122 in hepatocarcinoma cells,and analyze the relationship between the expression of miR-122 and cancer cell cycle regulation.Methods The in situ hybridization was used to detect the expression of miR-122 in 26 cases of primary hepatocarcinoma,and the flow cytometry was used to analyze the cancer cell cycle.Results The positive rate of miR-122 was 80.77% in liver tissues,and 26.92% in hepatocarcinoma(P0.01).The percentage of S-phase cells was increased and the percentage of G1/G0-phase cells was decreased in hepatocarcinoma compared to the liver tissue group(P0.05).In the miR-122 negative hepatocarcinoma group,the percentage of S-phase cells was increased and the percentage of G1/G0-phase cells was decreased obviously(P0.01),compared to the miR-122 positive hepatocarcinoma group.Conclusion Downregulation of miR-122 in hepatocarcinoma promotes the cancer cell division and makes the cancer cells have more proliferation,infiltration and metastatic abilities.
