清热滋阴活血复方对大鼠糖尿病牙周炎的治疗作用及外周血IL-1β的影响

摘　　要：目的通过动物实验,观察清热滋阴活血中药复方对大鼠糖尿病牙周炎的疗效,并初步探讨其机制。方法 10周龄健康雄性Wistar大鼠随机分为空白对照组(A组)、糖尿病牙周炎生理盐水组(B组)和中药治疗组(C组),B组与C组建立糖尿病牙周炎模型后,C组灌胃等效剂量的金银花、北沙参、川芎复方水煎剂,A组与B组灌胃等体积的生理盐水。2个月后,麻醉下方块截取包括实验区磨牙的双侧上颌骨。HE染色后,选择包括牙龈结缔组织及上皮组织的视野计数浸润的炎细胞,测量实验牙近中牙槽嵴顶至釉牙骨质界的距离;腹腔总静脉抽取静脉血,制备血清后用ELISA方法检测IL-1β的含量,应用SPSS10.0软件包对数据进行统计分析。结果 B组与C组牙龈组织的浸润炎细胞数目、牙槽嵴顶至釉牙骨质界的距离较A组均有增高,且B组高于C组,三组两两比较差异均具有统计学意义(P<0.05);B组外周血IL-1β的含量较A组与C组高,差异均有统计学意义(P<0.05)。结论清热滋阴活血复方对糖尿病牙周炎有治疗作用。Objective To observe the effect of the replenishing yin,clearing heat and promoting blood circulation compound prescription on the rats with diabetic periodontitis and to explore its mechanism.Methods 10-week-old healthy male Wistar rats were randomly divided into the blank control group（group A）,the diabetic periodontitis group（group B）and the traditional Chinese medicine treatment group（group C）.After diabetic periodontitis models were established in group B and C,group C was fed with equivalent dosage of Honeysuckle,North Adenophora and Chuanxiong compound decoction,and group A and B were fed with an equal volume of normal saline.2 months later,under anesthesia,boxes of the bilateral maxillary including experimental zone molar were intercepted.HE staining was used to detect general conditions and pathological changes by counting the number of infiltrating inflammatory cells in the gingival connective tissue and epithelial tissue and measuring the distance from the mesial alveolar crest to the cemento-enamel junction of the experimental tooth.Venous blood was drawn out from the main vein of the abdomen.After preparing the serum,the level of IL-1β was measured by ELISA and the data were statistically analyzed by SPSS10.0 softwarepackage.Results The number of infiltrating inflammatory cells in the gingival tissue and the distance from the alveolar crest to the cemento-enamel junction,were increased in groups B and C compared with those in group A,and there were significant differences among the three groups（P0.05）.Level of IL-1β in the peripheral blood of group B was higher than those of groups A and C,and there was a significant difference（P0.05）.Conclusion The replenishing yin,clearing heat and promoting blood circulation compound prescription has curative effect on diabetic periodontitis.
