rhIL-3增强急性髓系白血病细胞对柔红霉素敏感性的研究

摘　　要：目的观察经重组人白细胞介素３（ｒｈＩＬ３）刺激后急性髓系白血病（ＡＭＬ）细胞对柔红霉素（ＤＮＲ）敏感性的变化。方法用流式细胞仪、细胞形态学、联苯胺染色观察ｒｈＩＬ３对ＡＭＬ细胞增殖与分化的影响；用荧光分光光度法测定ＡＭＬ细胞内ＤＮＲ浓度；用半固体培养法观察ＣＦＵＡＭＬ产率。结果①ｒｈＩＬ３对体外培养的ＡＭＬ细胞具有促增殖作用，但无明显诱导分化作用；②ｒｈＩＬ３不影响ＡＭＬ细胞对ＤＮＲ的摄取与排泄平衡，但能增强ＡＭＬ细胞对ＤＮＲ的敏感性。结论ｒｈＩＬ３与ＤＮＲ联合应用，有可能提高临床治疗ＡＭＬ的疗效。Objective To evaluate the effect of recombinant human interleukin 3(rhIL 3) on the sensitization of acute myeloid leukemia(AML) cells to daunorubicin(DNR).Methods AML cells were cultured in a DNR containing medium supplemented or not with rhIL 3. CFU AML was assayed by semi solid culture, proliferation and differentiation of AML cells were assayed by flow cytometry, cytomorphology and benzidine staining.Intracellular DNR concentration was measured by spectrophotofluorometry.rhIL 3 could enhance the proliferation but not differentiation of AML cells in vitro, and had no effect on DNR intake and exclusion of AML cells while enhancing their sensitivity to DNR. Conclusion rhIL 3 combined with DNR might improve the therapeutic effect of AML.
