摘　　要：目的:探讨血管内皮生长因子(VEGF)在胃癌组织中的表达及其对基质金属蛋白酶-9(MMP-9)的表达的影响在胃癌侵袭机制中的意义。方法:采用免疫组织化学法、MTT法、实时定量PCR及Westernblot,观察胃癌组织及癌旁组织中VEGF的表达、VEGF对胃癌AGS细胞增殖和AGS细胞中MMP-9mRNA及蛋白表达的影响。结果:VEGF在胃癌中高表达;经VEGF刺激后,AGS细胞增殖明显,MMP-9mRNA及蛋白表达亦明显增加,具有统计学意义(P<0.05)。结论:VEGF通过上调MMP-9的表达促进胃癌的侵袭转移,VEGF和MMP-9可作为了解胃癌生物学行为和判断预后的指标。Objective： To investigate the expression of vascular endothelial growth factor （VEGF）and the significance of matrix metalloproteinases-9 （MMP-9）, which is modulated by VEGF, in the invasion of gastric carcinoma. Methods： Expression of VEGF in gastric carcinoma and normal gastric tissues was detected by Western blot. MTT was used to measure the proliferation of AGS cells while real-time PCR and Western blot were used to measure the expression of MMP-9 at mRNA and protein level in AGS cells. Results： The expression of VEGF in gastric carcinoma was significantly higher than that of controls; the expression of MMP-9 in AGS cells with VEGF was significantly higher than that of controls at mRNA and protein level. Conclusion： Both VEGF and MMP-9 participate in the progression of gastric carcinoma, and VEGF may promote the invasion of gastric carcinoma by upregulating the expression of MMP-9.
