慢性间歇性缺氧合并脑缺血再灌注动物模型的研究

摘  要:目的:通过慢性间歇性缺氧(CIH)合并脑缺血再灌注(CIR)大鼠模型的建立,研究睡眠呼吸暂停综合征(OSAS)对脑梗塞的影响。方法:2月龄Sprague-Dewley(SD)大鼠34只,随机选取10只在缺氧箱中行血氧检测,余随机分成4组:空白对照组(UC,n=6),假手术组(n=6),CIR组(n=6),CIH+CIR组(n=6)。CIH+CIR组大鼠每天间歇性缺氧8小时。UC组、假手术组及CIR组暂不作处理。8周后CIR组和CIH+CIR组均用线栓法制备大脑中动脉缺血(MCA0)模型。通过神经功能缺损评分观察大鼠大体情况;2,3,5-氯化三苯基四氮唑(TTC)染色观察脑梗死体积。结果:CIH+CIR组与CIR组梗死体积和神经功能评分均有明显的差异。结论:本实验建立的慢性间歇性缺氧合并脑缺血再灌注大鼠模型可满足OSAS+脑梗死基础研究的需要,CIH可加重脑缺血再灌注损伤。Obejective:To study the effect of obstructive sleep apnea syndrome(OSAS)on stroke by establishing the chronic intermittent hypoxia(CIH)combining cerebral ischemia ruperfusion(CIR)model in rat.Methods:thirty four two-month old male Sprague-Dawley(SD)rats were chosen.Ten rats were carried out randomly to be monitored arterial blood oxygen saturation in hypoxic box.The remained twenty four rats were randomly assigned four experimental groups:unhandled contrl group(UC,n=6)、sham operation group(n=6)、CIR group(n=6)、CIH combining CIR group(n=6).Rats in CIH group suffered 8 hours intermittent hypexia everyday,and those in UC group、sham operation group、CIH combining CIR group were raising normal.The model of middle cerebral artery occlusion(MCA0)was induced in CIR group and CIH combining CIR group with suture method after 8 weeks.The rats general situation was evaluated with neurological deficit score.infarction with 2,3,5-chloride triphenyltin tetrazolylazo (TTC)staining.Results:the infarction and neurological deficit score in CIH combining CIR group was significantly different from that in CIR group.Conclusions:The chronic intermittent hypoxia combining cerebral ischemia ruperfusion model in rat in our experiment can satisfy the need of fundamental study on OSAS combining stroke.CIH enhances cerebral ischemia ruperfusion injury.
