摘  要:目的探讨Behet病患者外周血单核细胞(peripheral blood mononuclear cell,PBMC)细胞因子信号抑制因子(sup-pressor of cytokine signaling,SOCS)表达以及SOCS在Behet病发病机制中的作用。设计病例-对照研究。研究对象16例Behet病患者及16例正常人。方法取Behet病患者和正常人的静脉血,用ELISA方法检测血清中细胞因子IL-4、IL-12、IFN-γ的水平;用实时荧光定量PCR和蛋白免疫印迹方法检测外周血单核细胞内SOCS基因和蛋白的表达。主要指标Behet病患者和健康人之间IL-4、IL-12、IFN-γ、CIS、SOCS1、SOCS2、SOCS3、SOCS5含量。结果 IL-4、IL-12和IFN-γ在Behet病患者血清中的水平均较正常对照组显著升高(P均<0.05)。相对荧光定量PCR结果显示含SH2结构的细胞因子诱导蛋白(cytokinein-ducible SH2 containing protein,CIS)、SOCS1、SOCS2、SOCS3、SOCS 5mRNA在Behet病患者PBMC内的表达分别是正常对照组的0.42、1.19、1.31、0.64和1.16倍。蛋白免疫印迹结果显示Behet病患者的CIS水平显著低于正常人(P<0.01),SOCS1明显高于正常人(P<0.01),SOCS2在患者和正常对照组之间无显著差异(P>0.05),SOCS3低于正常对照组(P<0.01),SOCS5高于正常对照组(P<0.01)。结论 Behet病患者PBMC内SOCS1和SOCS5水平升高、CIS和SOCS3水平降低与Behet病Th1型反应增强有关。SOCS5在Behet病发生过程中可能起保护作用。Objective To investigate suppressor of cytokine signaling (SOCS) mRNA and protein expression on peripheral blood mononuclear cells (PBMC) in patients with Behcet's disease with active uveitis, to explore the role of SOCS in the pathogenesis of Behcet's disease. Design Case-control series. Participants Sixteen Behcet's disease patients and 16 healthy individuals. Methods Blood samples were taken from 16 Behcet's disease patients with active uveitis and 16 healthy individuals. IFN-γ, IL-12, and IL-4 in the serum were measured by enzyme-linked immunosorbent assay (ELISA). PBMC were subjected to analyze of SOCS mRNA and protein expression using quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and western blot, respectively. Main Outcome Measures IL-4, IL-12 ,IFN-γ,CIS,SOCS1,SOCS2,SOCS3,SOCS5 were analyzed between Behcet's disease patients and healthy individuals. Results The level of IFN-γ, IL-12 and IL-4 were all significantly higher in the serum of patients than that in controls(P〈O.O1 ). Cytokine inducible SH2 containing protein (CIS) mRNA, SOCS1 mRNA, SOCS2 mRNA, SOCS3 mRNA and SOCS5 mRNA levels in PBMC of Behcet's disease patients with active uveitis were 0.42, 1.19, 1.31, 0.64 and 1.16 folds than that in healthy volunteers. Marked decreased expression of CIS protein was detected in active patients than that in controls (P〈O.01),SOCS1 protein was up-regulated significantly in active patients than that in controls (P〈0.01), there was no significant difference in SOCS2 between two groups,SOCS3 protein was significantly decreased in patients than that in controls (P〈0.01), SOCS5 protein was much higher in patients than in controls(P〈0.01 ). Conclusions Up regulation of SOCS1 and SOCS5 expression and down-regulation of SOCS3 and CIS may correlate with the development of Thl mediated immune response in Behcet's disease.
