摘  要:【目的】探讨大肠癌中转移相关基因1(MTA1)、缺氧诱导因子-1α(HIF-1α)蛋白的表达及其与淋巴管生成的关系。【方法】用免疫组织化学SP法检测81例大肠癌中MTA1、HIF-1α蛋白的表达及D2-40标记的淋巴管密度。【结果】81例大肠癌中MTA1、HIF-1α阳性表达率分别为66.7%和63%,MTA1和HIF-1α的表达均与大肠癌Duke's分期和淋巴结转移相关(P<0.05)而与组织学分级和浸润深度无关(P>0.05),大肠癌中MTA1和HIF-1α蛋白表达呈正相关(P<0.05),MTA1和HIF-1α蛋白过表达病例肿瘤旁淋巴管密度分别为18.4±4.9、18.0±5.3,高于MTA1和HIF-1α不表达和弱表达病例(15.6±4.5、15.4±4.2),两者的差异有统计学意义(P<0.05)。【结论】MTA1和HIF-1α蛋白的过表达可能促进了大肠癌旁淋巴管的生成,在肿瘤淋巴结的转移中起一定作用。【Objective】To investigate the expression of metastasis-associated gene 1(MTA1),hypoxia inducible-factor l alpha(HIF-1α)in human colorectal cancer and to explore the relationship between their expression and lymphangiogenesis.【Methods】The expression of MTA1 and HIF-1α,and D2-40 marked lymphatic vessel density(LVD)were detected by immunohistochemical staining in eighty-one human colorectal cancer cases.【Results】The positive rates of MTA1 and HIF-1α expression were 66.7%and 63.0%respectively in the eighty-one human colorectal cancer cases.The expression of MTA1 and HIF-1αwere both correlate with Duke's stage and lymph node metastasis(P0.05)but did not correlate with the pathological grade and depth of invasion(P0.05).In addition,MTA1 and HIF-1αexpression has a positive correlation in human colorectal cancer(P0.05).In MTA1 and HIF-1αover-expression cases,the peritumoral LVD was 18.4±4.9 and 18.0±5.3 respectively and was higher than that of the MTA1 and HIF-1αexpression none or weak cases(15.6±4.5,15.4±4.2),the difference has statistically significance(P0.05).【Conclusion】The overexpression of MTA1 and HIF-1αin colorectal cancer possibly promote the cancer lymphangiogenesis and play a role in lymph node metastasis.
