摘 要:目的探讨核转录因子-κB(NF-κB)在纤维化肝脏枯否细胞活化中的作用机制。方法 Wistar大鼠随机分为正常对照组、肝纤维化模型组和吡咯烷二硫代氨基甲酸盐(PDTC)组,PDTC组大鼠在给CCl4的同时给予PDTC灌胃。HE染色,肝组织羟脯氨酸(Hyp)含量测定,赖氏法测定丙氨酸氨基转移酶(ALT),TBA法检测丙二醛(MDA)的水平,免疫组化法测定NF-κB和ED2的表达。结果免疫组化结果显示NF-κBp65在正常肝组织中少量表达于中央静脉周围的肝细胞胞浆内,纤维化模型组肝组织可见程度不等的细胞浆和(或)细胞核表达,主要分布于纤维化区、肝窦区、血管壁,部分胆管细胞、变性肝细胞也可见阳性染色,阳性细胞数从第1周末就显著高于正常对照组,PDTC组阳性细胞明显比同期纤维化模型组减少(P<0.05);ED2表达在正常对照组沿肝血窦少量散在分布,纤维化模型组阳性信号明显增多,主要分布在增生的纤维组织及纤维间隔,在肝实质内也有少量分布,阳性信号随着实验的进程呈递增趋势,且阳性细胞胞浆丰富、体积变大,外形为星形、多角形和不规则形。PDTC组阳性细胞数明显比同期纤维化模型组减少(P<0.05)。与正常对照组比较,纤维化模型组血浆ALT和肝组织MDA、Hyp含量从第1周末起明显增高,ALT和MDA呈先增高后降低趋势,但第6周末仍高于正常对照组,Hyp含量呈递增趋势;PDTC组ALT、MDA、Hyp比同期纤维化模型组明显降低。结论 NF-κB是纤维化肝脏枯否细胞活化的关键因子。Objective To study the effect of nuclear factor κB(NF-κB)on kuffer cells(KCs)activation during experimental hepatic fibrosis.Methods Wistar rats were randomly divided into normal control group,hepatic fibrisis model group and the Pyrrolidine-1-dithiocarboxylic acid ammonium sail(PDTC)group.The PDTC group was treated with subcutaneous injection of carboan tetrachloride,and treated with PDTC by oral administration at the same time.The content of hydroxyproline was measured.The alanine aminotransferase(ALT)of plasma was measured by laishi method.The content of malondialdehyde(MDA)in liver tissue was determined by TBA method.The expressions of NF-κB and ED2 were determined by immunohistochemistry.Results In control goup,just a small amount of NF-κB p65 was expressed in the cytoplasm of a few hepatocytes around central veins.In model goup,the positive staining of NF-κB p65 was expressed in cytoplasm and nucleus,mainly in fibrous stepta,hepatic sinusoid and partial vascular endothelial cells,part of proliferating ductular epithelial cells and impaired hepatocytes.The positive cells were obviously more in model group than those in contral group from the first week.The expression of NF-κB in liver tissues in PDTC groups was lower than that in model control groups(P0.05).A small amount of positive staining of ED2 was visible around fibrous septa.However,compared with control group,the expression of ED2 signifcantly was up-regulated mainly in fibrous septa.It was also detected in a few heptic cells.Futhermore,the positive staining cells cytoplasm was affluent and the volume became larger.They took the shape of star or polygon,and some were irregular-shaped.The expression of ED2 in liver tissues was lower than that in model and control groups at the corresponding time period in PDTC groups.Compared with normal control group,the levels of ALT and MDA and Hyp significantly increased in hepatic fibrosis group.Compared with the model group,the levels of ALT and MDA and Hyp decreased in different de
